双重阻断肾素血管紧张素系统对糖尿病肾病大鼠肾小管间质纤维化的影响

目的观察双重阻断肾素血管紧张素系统（RAS）对糖尿病肾病大鼠肾小管间质TGF-β1、CTGF、FN表达的影响。方法将STZ诱导的糖尿病肾病大鼠模型随机分为对照组（A组）、贝那普利组（B组）、贝那普利联合氯沙坦组（C组）。B组给予贝那普利10mg／kg灌胃，C组给予贝那普利5mg／ks、氯沙坦10mg／kg灌胃，8周后免疫组化观察各组肾小管间质TGF-β1、CTGF、FN表达，RT-PCR测定TGF-β1、CTGF、FN的mRNA表达。结果B组、C组大鼠小管间质TGF—β1、CTGF、FN表达、mRNA表达较A组均明显下调（P〈0．01），同时C组TGF-β1、CTGF、FN表达较B组也显著下调（P〈0．05）。结论贝那普利与氯沙坦联合阻断RAS，比单用贝那普利能更有效下调糖尿病大鼠肾小管间质TGF-β1、CTGF、FN的表达。

Effect of Dual Blockade of Renin Angiotensin System on Tubulointerstitial Fibrosis in Diabetic Nephropathy Rats

Objective To investigate the effect of dual blockade of rennin angiotensin system on tubulointerstitial fibrosis in diabetic nephropathy rats. Methods 60 STZ-induced diabetic nephrop- athy tats models were randomly divided into three groups： Normal control group （A group）, group with Benazepril therapy（ 10 mg/kg B group）, group with Benazepril （5 mg/kg）and Losartan （10 mg/kg）therapy （C group） ;At eight weeks after therapy, the expression of TGF-β1, CTGF and FN in tubulointerstitial were detected by immunohistochemistry and RT-PCR. Results Compared with A group and B group, the expression of TGF-β1 ,CTGF and FN were significantly down-regulated （P〈0.05 or P〈0. 01） in tubulointerstitial of C group. Conclusion Dual blockade of RAS with intermediate dose ACEI and ARB is more effective in down-regulating the expression of TGF-β1 ,CTGF and FN in tubulointerstitial of diabetic nephropathy rats.
