碘化钾对实验性铅性肾病的保护作用及其对转化生长因子-β表达的影响

摘    要:目的:观察碘化钾对铅性肾病的保护作用,并从蛋白质和分子水平观察其对转化生长因子-β(TGF-β)表达的影响。方法:SD大鼠48只(180~220 g)适应性饲养1周,随机分为两组,每组根据时间点的不同又分3个亚组。APb,BPb,CPb为染铅组,分别给予0.5%的醋酸铅喂饮1,2,3个月;AKI,BKI,CKI为碘化钾组,给予0.5%醋酸铅喂饮的同时予碘化钾(3 mg/100 g体重),用注射用水稀释灌胃每日1次,时间分别为1,2,3个月,所有实验组中大鼠每天的饮水量大致相同。实验结束时称体重、肾重;原子吸收光谱石墨炉法测量血铅、肾铅含量;用免疫组织化学的方法检测肾组织TGF-β蛋白的表达,用RT-PCR的方法检测肾组织TGF-βmRNA的表达。结果:实验到第3个月时,碘化钾组与染铅组比较体重差显著增高,肾重/体重比显著降低;血铅、肾铅含量显著降低;而实验前2个月无明显差别。肾组织TGF-β蛋白和mRNA的表达在两组中差异无显著性。结论:碘化钾能拮抗铅对大鼠生长发育的影响,并有驱铅的作用,但对TGF-β的影响不大。

Abstract:Objective:To investigate the protective effect of potassium iodide on experimental lead nephropathy,and its influence on the expression of transforming growth factor-βin kidney at molecular.Methods:Forty-eight SD rats weigh 180~220 g were randomly assigned into two groups.The rats in Pb groups were given 0.5% lead acetate water for 1,2,3 months respectively(APb,BPb,CPb),while the rats in KI groups were given 0.5% lead acetate water simultaneously taking potassium iodide 3 mg/100 g body weight by intragastric administration for 1,2,3 months respectively(AKI,BKI,CKI).All rats were sacrificed at the end of the treatment.The body and renal weigh were measured.Lead concentrations in blood and kidney were determined by atomic absorption photometry with non-flame graphite oven.Expression of TGF-βprotein and mRNA in kidney was measured by immunohistochemistry and RT-PCR respectively.Results:The weight gain in CKI group was significantly increased as compared with CPb group,while the ratio of kidney weight/body weight.Blood and renal lead concentrations were significantly decreased as compared with CPb group.The expression of TGF-βprotein and mRNA in all groups had no significant difference.Conclusion:Potassium iodide could antagonize the lead-induced reduction in growth and lustrate lead,while it has no effect on TGF-βexpression in this study.
