摘    要:目的观察HMGB1在大鼠小肠缺血再灌注损伤中的表达及其意义。方法建立大鼠小肠缺血再灌注模型,随机分为Sham组、I/R组、I/R+A—box组,采用HE法检测大鼠小肠组织损伤程度,ELISA法检测大鼠血浆HMGB1、SOD、MDA浓度,免疫印迹法检测小肠组织HMGB1蛋白表达。结果I/R组血浆及肠组织HMGB1增加,血浆SOD浓度下降。MDA浓度升高,大鼠肠组织损伤加重;I/R+A—box组血浆及肠组织HMGB1表达较I/R组下降,血浆SOD浓度升高,MDA浓度下降,大鼠肠组织损伤减轻,HMGB1-Abox具有保护作用。结论HMGB1在肠缺血再灌注后表达增加,组织氧化程度加重.组织损伤加重,提示HMGB1可能是肠缺血再灌注损伤的独立危险因子。

Expression and its significance of HMGB1 in intestinal ischemia reperfusion injury of rats

Abstract:Objective To explore the changes and significance of HMGB1 in the intestinal ischemia reperfusion injury of rats.Methods Intestinal ischemia reperfusion injury model in rats were made and randomly divided into sham group,I/R group and I/R+A-box group.Intestinal tissue injury severity of rat was detected by HE assay.The HMGB1,SOD and MDA concentrations in plasma were measured by ELISA,and HMGB1 protein expression in intestinal tissue was detected by immunoblot assay.Results The expression of HMGB1 significantly increased in the ischemic area in I/R group compared with sham group,while SOD concentration decreased,MDA concentration elevated,and rat intestinal tissue injury was more serious.But intestinal tissue injury reduced in I/R+A-box group while HMGB1-A box had a protective effect.Conclusion The expression of HMGB1 in intestinal ischemia reperfusion increases.Tissue oxidation degree and tissue injury are serious.It suggests that HMGB1 may be an independent risk factor in intestinal ischemia-reperfusion injury.
