Abstract:Objective: To investigate the effect of dendritic cells (DCs) and regulatory dendritic cells (CD11c ^low CD45RB^high DCs) on thermal injury-induced inflammatory cytokine production through single intraperitoneal injection of DCs and CD11c^low CD45RB^high DCs to scald mice. Methods: Mice were subjected to a 15% total body surface area (TBSA) scald injury on the back. Both sham- and burn-injured mice received 1.0 ml of Ringer's solution for resuscitation. CD11c^low CD45RB^high DCs and DCs was added to lactated Ringer's solution (1 ml/mouse) in treatment groups. Animals of all groups were sacrificed at post burn hours (PBH) 12, 24 and 48 respectively, and peritoneal fluid samples were harvested aseptically and stored immediately at - 70 ℃ until use for measurement of cytokine levels. Spleens were used to procure CD11c^low CD45RB^high DCs and DCs. Result: IL-6 levels in peritoneal fluid in CD11c^low CD45RB^high DCs group (296.57±11.40), (152.89±6.03), (53.18±3.25) pg/ml] and DCs group (602.44±20. 64), (304.35±9.46), (118.30±8.90)pg/ml] were significant decreased compared with scald group (896.21±24.63) ,(451.74±13.22) ,(199.02±7.83) pg/ml] at PBH 12, 24 and 48 (all P〈0. 01). Similarly, MCP-1 levels in peritoneal fluid in CD11c^low CD45RB^high DCs group (1 308.40±87.90), (472.00±29.37), (61.29±5.20) pg/ml] and DCs group (1 708.36±103.22), (789.23±49.02), (86.33±6.11) pg/ml] were markedly lowered compared with untreated mice (2 019.59± 129.66), (1 067.90±65.88), (152.87± 14.28) pg/ml] at PBH12, 24 and 48 (all P〈0.01). Moreover, TNF-a levels in peritoneal fluid in both CD11c^low CD45RB^high DCs and DCs treated group were significantly lower than that in untreated mice (81.63 ±6.02), (54.74±4.01), (29.45±2.51) pg/ml] at PBH 12, 24 and 48 (P〈0.01). In addition, IL-6, MCP-1 and TNF- a levels were markedly lowered in CD11c^low CD45RB^high DCs group compared with DCs group at various intervals (P 〈0.01) Conclusion: Naturally existed CD11c^low CD45RB^high DCs and DCs might effectively inhibit excessive inflammation in mi ce with major burns.
