c-myc在消化系统肿瘤的表达及其与端粒酶活性关系的研究

摘    要:为了解c-myc在消化系统肿瘤的表达以及c-myc与端粒酶活性的关系,采用链酶抗生物素蛋白-碱性磷酸酶免疫组织化学技术对67例消化道肿瘤及15例消化道良性病变的c-myc蛋白表达进行研究,并采用方差分析及相关系数对其进行统计学处理,结果各组织标本均有不同程度的c-myc蛋白表达,肿瘤组与非肿瘤组c-myc双样本方差分析P>0.05;不同组织间c-myc双样本方差分析P<0.05,低分化癌与高分化癌及中分化癌统计结果分别为P<0.05,P<0.001,c-myc表达与端粒酶活性的相关系数=0.31,端粒酶阳性与阴性组c-myc表达P>0.05,但端粒酶阳性组c-myc表达平均积分高于阴性组,结果表明:(1)随着肿瘤分化程度的减低c-myc蛋白表达减弱;(2)c-myc蛋白表达可能为组织增生的标志并非肿瘤特异性标志物;(3)c-myc蛋白表达与端粒酶活性有一定的关系。

Abstract:Objective : to understand the expression of c-myc in the digestive system tumors and the relationship between c-myc and telomerase. Methods: the expression of c-myc were determined by immunohistochemical SP method in specimens of the digestive system tumors (67 cases) and in the benign ones (15 cases) . Meanwhile the analysis of data was made by F-test and correlation index. Results: all specimens showed various degrees of P 62c-myc staining; the expression of c-myc between tumor group and benign one has no significance(P>0 05); variation in carcinomas staining with myc9E/11 was demonstrated considerably depending on histological grade. Staining in poorly differentiated tumors was far higher than in both the well and the middle ones(P<0 05, P<0 001 respectively); there was a positive correlation between c-myc expression and activity of telomerase( r =0 38);although the expression of c-myc in the both activated and unactivated telomerase groups(P>0 05), the expressive average of c-myc in the positive was higher than in the negative. Conclusion: c-myc 9E/11 protein was located in both nuclear and cytoplasm; the expression of c-myc was a marker of cellular proliferation but not of malignancy; the expression of c-myc was related with the tumor differentiation and with the telomerase activity.
