硝酸镧对大鼠脾脏形态及脾细胞IL-2和IFN-γ分泌水平的影响

摘    要:目的:研究不同剂量硝酸镧作用不同时间对大鼠脾脏形态及机能的影响.方法:取大鼠脾脏进行脾体比、常规组织学染色观察,同时取脾细胞进行淋巴细胞转化率试验及IL-2与IFN-γ的测定.结果:不同剂量硝酸镧灌胃1个月后脾体比、脾脏形态结构及免疫功能与对照组相比差异无显著性;不同剂量硝酸镧灌胃6个月后脾体比及脾脏形态结构与对照组相比差异无显著性,但20 mg@kg-1的硝酸镧使脾淋巴细胞转化率下降,IL-2和IFN-γ水平有所下降,而10.0、2.0、0.2和0.1 mg@kg-1硝酸镧剂量组均可使大鼠脾淋巴细胞转化率升高,分泌IL-2和IFN-γ水平上升.结论:不同剂量硝酸镧灌胃1个月对大鼠脾脏形态及免疫功能无明显影响;20 mg@kg-1的硝酸镧灌胃6个月使大鼠脾脏细胞免疫功能下降,而10.0、2.0、0.2和0.1 mg@kg-1的硝酸镧连续灌胃6个月则增强了大鼠脾脏细胞免疫功能.

Abstract:Objective: To study the effect of different doses of lanthanum nitrate La(NO 3) 3] on morphology and function of spleen in rats.Methods: Ratios of spleen to body weight were measured. Light microscopy and lymphocyte transformation test were used, and the levels of IL 2 and IFN γ of splenic lymphocytes were detected in this study. Results: Compared with the control group, ratios of spleen to body weight, the morphology and immunologic parameters of spleen were not varied in experimental groups after administration of La(NO 3) 3 for a month. After taking La(NO 3) 3 orally for 6 months, ratios of spleen to body weight and the morphology of spleen were not different compared with control group; the transformation rates of lymphocytes and IL 2 and IFN γ levels were decreased in the group of 20 mg·kg -1 La(NO 3) 3 . The transformation rates of lymphocytes and the IL 2 and IFN γ levels were increased in the groups of 10.0, 2.0, 0.2 and 0.1 mg·kg -1 La(NO 3) 3. Conclusion: Cell mediated immunity was suppressed in the group of 20 mg·kg -1 La(NO 3) 3 . But promoted in the groups of 10.0 ,2.0, 0.2 and 0.1 mg·kg -1 La(NO 3) 3 after taking La(NO 3) 3 orally for 6 months.
