己酮可可碱对大鼠脑缺血再灌注损伤细胞凋亡及Bcl-2、Caspase-3表达的影响

摘    要:目的:探讨己酮可可碱对大鼠局灶性脑缺血再灌注损伤Bcl-2、Caspase-3表达的影响。方法:sD大鼠随机分为假手术组、脑缺血-再灌注模型组、川芎嗪组和己酮可可碱组,每组10只。术前实验性灌胃给药,川芎嗪给药剂量为50mg/kg,fyllX组给药剂量为100mg/kg,每日-次,连续给药10天。采用线栓法制作大鼠脑缺血-再灌注损伤模型。造模成功2h后再尾静脉注射1次。假手术组和对照组给予生理盐水。用TUNEI.法检测脑缺血2h再灌注24h大鼠皮质和海马神经细胞凋亡率,免疫组化法观察Bcl-2和Caspase-3蛋白表达。结果:与脑缺血-再灌注组比较,川芎嗪组和PTX组皮质和海马部神经细胞凋亡率明显减少(P均〈0.01),Bcl-2表达明显增加(P均〈0.01),Caspase-3表达明显降低(P均〈0.01),川芎嗪组和Prx组两组间比较差异无统计学意义(P〉0.05)。结论:PTX可抑制大鼠神经细胞凋亡,促进Bcl-2表达,降低Caspase-3表达。
