佐剂性关节炎大鼠血小板活化与FAK/Calpain信号通路紊乱有关

摘    要:目的:探讨佐剂性关节炎大鼠血小板活化与FAK/Calpain信号通路的关系。方法:将20只大鼠随机分为正常对照(NC)组、模型对照(MC)组,每组10只,向模型组动物右后足跖皮内注射弗氏完全佐剂0. 1 ml致炎。致炎30 d后,观察各组大鼠体重(M)、足跖肿胀度(E)、关节炎指数(AI)、关节病理变化、FAK/Calpain信号通路、细胞因子(IL-6、IL-8、MMP-9)、血小板参数及血小板活化相关指标(CD40L、CD147、GPⅡb/Ⅲa)的变化。结果:与NC组相比,MC组大鼠E、AI、CD40L、CD147、GPⅡb/Ⅲa、FAK、p-FAK、IL-6、IL-8、MMP-9、PLT、PCT表达明显升高(P <0. 01); Calpain1、Calpain2表达明显降低(P <0. 01)。相关性分析显示FAK蛋白与CD147、GPⅡb/Ⅲa呈正相关,p-FAK蛋白与CD40L呈正相关,Calpain1蛋白与IL-6呈负相关,Calpain2蛋白与E呈负相关,FAK mRNA与IL-6呈正相关。结论:AA大鼠不但存在关节局部的炎症反应,而且血小板也存在活化现象,其机制可能与FAK/Calpain信号传导通路的变化及细胞因子的紊乱有关。

Abstract:Objective:To investigate the relationship between platelet activation and FAK/Calpain signal pathway in adjuvant arthritis rats.Methods:A total of 20 rats were randomly divided into a normal group and a model group.Rats in the model group were induced to establish the adjuvant arthritis AA model by intradermally injecting 0.1 ml Freund′s complete adjuvant into the right paw.After 30 days,the paw edema volume,arthritis index,and the changes of joint pathological,FAK/Calpain,IL-6,IL-8,MMP-9,platelet parameter and platelet activation correlation indexes(CD40L,CD147,GPⅡb/Ⅲa)were observed.Results:Compared with the normal group,the paw edema volume,arthritis index,CD40L,CD147,GPⅡb/Ⅲa,IL-6,IL-8,MMP-9,PLT,PCT and FAK,p-FAK of the synovial tissue in the model group was significantly increased,while Calpain1 and Calpain2 of the synovial tissue in the model group was significantly decreased.There were significant positive correlations between FAK protien and CD147,GPⅡb/Ⅲa,p-FAK protien was significant positive correlated with CD40L,FAK mRNA was significant negative correlated with IL-6,while Calpain1 protien was significant negative correlated with IL-6,Calpain2 protien was significant negative correlated with E.Conclusion:The mechanism of joint local inflammation and platelet activation may be related to the change of FAK/Calpain signaling pathway and the disorder of cytokines in AA rats.
