虫草素对胶质瘤细胞的增殖及侵袭作用的影响及机制研究

摘    要:目的:探讨虫草素对胶质瘤细胞增殖及侵袭作用的影响,并初步探讨其机制。方法:将虫草素配置成不同浓度(100、200和400μg/ml)作用于胶质瘤细胞,对照组不加入药物。使用CCK8法检测细胞不同时间点和不同药物浓度增殖情况,采用Transwell实验检测不同药物浓度作用下细胞侵袭情况,通过Westernblot和q-PCR法检测CyclinD1、Bcl-2、MMP-2、MMP-9、Ezrin蛋白和mRNA的表达水平。结果:100μg/ml虫草素对U87细胞增殖和侵袭无明显影响(P>0.05)。200和400μg/ml虫草素可明显抑制U87细胞增殖,呈剂量和时间依赖性(P<0.05)。200和400μg/ml虫草素可明显抑制U87细胞侵袭,呈剂量依赖性(P<0.05)。U87细胞经不同浓度虫草素(100、200、400μg/ml)处理24h后,发现200和400μg/ml虫草素处理后CyclinD1、Bcl-2、MMP-2、MMP-9、Ezrin蛋白和mRNA水平明显降低(P<0.05),100μg/ml虫草素处理后对CyclinD1、Bcl-2、MMP-2、MMP-9、Ezrin蛋白和mRNA水平的抑制作用不明显(P>0.05)。结论:虫草素能抑制U87细胞的增殖和侵袭,其机制可能与下调CyclinD1、Bcl-2、MMP-2、MMP-9、Ezrin的表达有关。

Abstract:Objective: To investigate the effect of cordycepin on the proliferation and invasion of glioma cells and to explore its mechanism. Methods: Cordycepin was dosed at different concentrations (100,200 and 400 μg/ml) to the glioma cells,and no drug was added to the control group.CCK8 method was used to detect cell proliferation at different time points and different drug concentrations.Transwell assay was used to detect the effect of different drug concentrations on cell invasion.The expression levels of Cyclin D1,Bcl-2,MMP-2,MMP-9 and Ezrin protein and mRNA were detected by Western blot and q-PCR. Results: The concentration of 100 μg/ml cordycepin had no significant effect on the proliferation and invasion of U87 cells ( P >0.05).U87 cell proliferation was significantly by inhibited 200 and 400 μg/ml cordycepin,and it was in a time- and dose-dependent manner( P <0.05).The U87 cell invasion was inhibited in 200 and 400 μg/ml cordycepin,and it was in a dose-dependent manner( P <0.05).U87 cells were treated with different concentrations of cordycepin (100,200,400 μg/ml) for 24 h,the levels of Cyclin D1,Bcl- 2,MMP-2,MMP-9 and Ezrin protein and mRNA were significantly decreased after treatment with 200 and 400 μg/ml cordycepin ( P <0.05).The inhibition of Cyclin D1,Bcl-2,MMP-2,MMP-9 and Ezrin protein and mRNA levels after treatment with 100 μg/ml cordycepin was not obvious ( P >0.05). Conclusion: Cordycepin can inhibit the proliferation and invasion of U87 cells,and its mechanism may be related to the down-regulation of Cyclin D1,Bcl-2,MMP-2,MMP-9 and Ezrin expression.
