消银汤治疗银屑病小鼠模型的实验研究

摘    要:目的:通过检测消银汤对银屑病小鼠增殖细胞核抗原(PCNA)、血管内皮生长因子(VEGF)和血小板内皮细胞黏附分子(CD31)的影响,探讨消银汤对银屑病小鼠模型的治疗作用。方法:BALB/c雌性小鼠50只,随机分为对照组、模型组、消银汤高、中和低剂量组,每组10只。实验前1 d全部小鼠背部皮肤去毛(3 cm2);次日模型组和消银汤各剂量组小鼠背部去毛处皮肤均匀涂抹10%十二烷基硫酸钠(SDS) 2 ml,1次/d,共7 d制备模型;对照组小鼠给予蒸馏水同样涂抹7 d。消银汤组采用灌胃方式给予17. 3、34. 5和69. 0 g/kg的消银汤,每天2次,连续14 d,对照组及模型组均每日给予等剂量的生理盐水,然后处死取血液和皮肤组织。采用苏木精-伊红(HE)染色观察小鼠皮肤组织的病理改变;采用实时荧光定量PCR法和免疫组织化学方法检测小鼠血液和皮肤组织中PCNA、VEGF和CD31的mRNA水平及蛋白的阳性表达强度。结果:HE染色结果显示,与对照组比较,模型组小鼠皮肤表皮明显增厚,消银汤各剂量组小鼠皮肤表皮略增厚;模型组小鼠血液及皮肤组织中PCNA、VEGF和CD31的mRNA水平及蛋白的阳性表达强度均显著升高,差异有统计学意义(P<0. 05);与模型组比较,消银汤组小鼠皮肤组织中PCNA、VEGF和CD31的mRNA水平及蛋白的阳性表达强度均明显降低有统计学意义(P<0. 05)。结论:消银汤能抑制银屑病小鼠表皮细胞增殖,明显改善银屑病小鼠的临床症状,可能与下调皮损处PCNA、VEGF和CD31的mRNA及蛋白表达水平有关。

Abstract:Objective:To investigate the effects of Xiaoyin decoction on PCNA,vascular endothelial growth factor(VEGF)and platelet endothelial cell adhesion molecule(CD31)in psoriatic mice,to explore the therapeutic effect of Xiaoyin decoction on psoriatic mice model.Methods:50 BALB/c female mice were randomly divided into control group,model group,high,medium and low dose groups with 10 rats in each group.On the first day before the experiment,all the mice were dehairing(3 cm 2)on the back,the next day,mice in model group and each dosage group of Xiaoyin decoction were evenly smeared with 10%sodium dodecyl sulfate(sodium dodecyl sulfate SDS)once a day for 7 days.And the mice in the control group were given the same amount of distilled water for 7 days.Xiaoyin decoction group was treated with 17.3 g,34.5 and 69.0 g/kg Xiaoyin decoction by gavage,two times a day for 14 days,the control group and the model group were given the same dose of saline daily,then killed to take blood and skin tissue.Hematoxylin eosin(HE)staining was used to observe the pathological changes of skin tissue in mice.The mRNA levels of PCNA,VEGF and CD31 in blood and skin tissues of mice were detected by real-time fluorescence quantitative PCR and immunohistochemistry.Results:The pathological results of HE staining showed that,the skin epidermis of the model group was thicker than that of the control group,and the skin epidermis of each dose group of Xiaoyin decoction was slightly thickened.In the model group,the mRNA levels and the positive expression of protein of PCNA,VEGF and CD31 in blood and skin were significantly increased(P<0.05),compared with the model group,the mRNA levels and the positive expression of protein of PCNA,VEGF and CD31 in blood and skin of mice in the Xiaoyin decoction group were significantly lower than those in the control group(P<0.05).Conclusion:Xiaoyin decoction can inhibit the proliferation of epidermal cells in psoriatic mice and improve the clinical symptoms of psoriatic mice,which may be related to the down-regulation of mRNA and protein expression of PCNA,VEGF and CD31 in psoriatic lesions.
