Influences of Actinomycin 23 21 on the Macromolecules in Sensitive Tumor Cells

Objective To study the antitumor activity of actinomycin 23 21 and the relation with macromolecules. Metheds The influences in lifespan were determined in mice with P388 and L1210 leukemia. In vitro antitumor activities were investigated by means of neutral red vital staining and trypan blue dye exclusion. The radiolabeled precursors incorporation was assessed by liquid scintillation counter. The intracellular cAMP quantity was determined by radioimmunoassay. Results Act23 21 produced significant inhibition in mice leukemias (in vivo) and in human tumor cells(in vitro). Optimal treatment regimen resulted in an increase in lifespan of more then 115% in mice with P388 leukemia. The tumor cell lines sensitive to Act23 21 include HL 60,SGC 7901,FGC85,HeLa and MGC 803,which is arranged in order of sensitivity. 3H UR incorporations were quickly and significantly inhibited in Act23 21 treated P388,SGC 7901 and HL 60 cells. The cAMP quantity enhenced significantly in Act23 21 treated EAC and HL 60 cells. Conclusion Actinomycin 23 21 possesses antitumor activity that is accompanied by the inhibition of RNA synthesis and enhance of cAMP quantity in tumor cells.
