香菇多糖对糖尿病大鼠巨噬细胞脂质过氧化和一氧化氮的影响

摘    要:目的:研究香菇多糖(LNT)对糖尿病大鼠巨噬细胞脂质过氧化作用和一氧化氮水平的影响。方法:SD大鼠分三组,正常对照组、糖尿病组、LNT治疗组,分别灌洗大鼠腹腔及肺泡巨噬细胞,并测定巨噬细胞内超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH—PX)、一氧化氮合酶(NOS)的活性及一氧化氮(NO)、丙二醛(MDA)的含量。结果:糖尿病大鼠腹腔及肺泡巨噬细胞内SOD、GSH-PX、NOS活性及NO含量下降,MDA含量增高,经LNT治疗后,SOD、GSH-PX、NOS活性及NO含量升高,MDA含量下降。结论:LNT可提高糖尿病大鼠巨噬细胞抗脂质过氧化作用及NO水平。

Abstract:Objective:To investigate the effect of lentinan on the lipid peroxidation and NO level in the macrophage from diabetic rats. Methods :Sprague-Dauley rats were divided into three groups; normal control group,diabetic group and lentinan treatment group,peritoneal and alveolar macrophage were harvested respectively from different group, the activity of superoxide dismutase(SOD) , glutathione peroxidase (GSH-PX), nitric oxide synthaae (NOS) and content of malondialde-hyde (MDA), nitric oxide (NO) were detetcted biochemically in the macrophage. Results: the activity of SOD, GSH-PX, NOS and the content of NO decreased while the content of MDA increased in the diabetic macrophage compared with control group,the activity of SOD, GSH-PX,NOS and the content of NO became increased but the content of MDA decreased in lentinan treatment group compaired with the diabetic one. Conclusion:Lentinan can enhance the capacity of anti-lipid per-oxidation and NO level of macrophage from diabetic rats.
