摘    要:为快速、有效地分析经空间飞行的蕃茄基因组DNA,采用了随机引物扩增多态DNA分析的分子标记方法,检测了基因序列的多态性。蕃茄种子搭载返地卫星,空间飞行355h,回收后地面种植,选择三个同工酶有差异的空间处理植株进行RAPD检测。结果表明:在50个供试引物中,有18个扩增出了DNA带,共有156条,其大小在200 ̄2000bp之间。

The Utility of RAPD Markers for Detection of Tomato DNA Mutation Induced by Space Flight.
Abstract:In order to investigate the change of tomato DNA after space flight,random amplified polymorphic DNA (RAPD)was used.It is a new method for molecular marking developed by Williams et al. in 1990.The dry seeds of tomato were carried on board the recoverable satellite,and then they were planted on the ground.Three individuals were used for RAPD analysis.In the RAPD reactions, 50 primers were used.18 primers among them amplify DNA fragments,varying from 200bp to 2000bp.Totally,5 primers showed polymorpic amplification products.The rates of DNA mutation of these three individuals tested are 4.5 %, 1.3 % and 3.2%, respectively.
