银杏叶提取物的HPLC/UV指纹图谱研究

摘    要:目的:建立银杏叶提取物的HPLC/UV指纹图谱。方法:以银杏叶提取物的HPLC/UV指纹图谱为参照物,采用HPLC法分析,并加以计算机辅助相似性评价系统对指纹图谱进行了相似度分析。采用Diamonsil(C18,250 mm×4.6mm,5μm)色谱柱;以流动相(A)水-乙腈-异丙醇-柠檬酸(1 000∶470∶50∶6.08)、流动相(B)水-乙腈-异丙醇-柠檬酸(1 400∶200∶30∶6.88)进行梯度洗脱;流速:1.0 ml/min,检测波长:360 nm。结果:在本试验条件下进行测试,可以建立银杏叶提取物以及银杏叶指纹图谱检测方法,得到分离度、重复性均较好的银杏叶提取物HPLC/UV指纹图谱,标示了13个共有峰,可以方便简单、并且有效控制不同厂家来源的银杏叶提取物。结论:银杏叶提取物指纹图谱的建立,为其质量控制提供了依据,也为银杏叶提取物以及银杏叶的指纹图谱国家标准的确立提供了更为科学的依据和有效的鉴别方法。

Abstract:Objective:To establish the fingerprints chromatograms of Ginkgo biloba extract by HPLC/UV.Methods:Ginkgo biloba extract with HPLC/UV fingerprint as the reference method using HPLC analysis and to computer-aided similarity evaluation system for the similarity analysis of fingerprints.Separation was performed of Diamonsil(C18,250 mm×4.6 mm,5 μm).Gradient elute was performed by the mobile phase consisting of water-acetonitrile-Isopropanol-citric acid(1 000∶470∶50∶6.08) and water-acetonitrile-Isopropanol-citric acid(1 400∶200∶30∶6.88) with the flow rate of 1.0 ml/min.The detecting wave length was set at 360 nm.Results:Perfect fingerprints chromatograms were obtained which can be used for the evaluation of Ginkgo biloba extract,13 common peaks were confirmed in fingerprints chromatograms.Conclusion:Ginkgo biloba extract Fingerprinting,provides a basis for quality control,but also for Ginkgo biloba extract and Ginkgo biloba fingerprint to establish national standards to provide a more scientific basis and effective method of identifying.
