桑生除痹合剂对膝骨性关节炎兔组织形态学和血液流变学的影响

目的：观察桑生除痹合剂对膝骨性关节炎家兔组织形态学和血液流变学的影响。方法：选取健康6月龄新西兰大白兔36只,将其随机分为正常组、造模组、对照组、桑生除痹合剂高（治疗Ⅰ组）、中（治疗Ⅱ组）、低（治疗Ⅲ组）剂量组,每组6只。按照Hulth造模方法造模,治疗Ⅰ组、治疗Ⅱ组、治疗Ⅲ组分别灌服高、中、低剂量桑生除痹合剂,对照组灌服壮骨关节丸。12周后,处死动物,取软骨及滑膜标本行光镜观察,并测定血液流变学指标。结果：关节软骨及滑膜标本光镜下呈显著退变表现;对照组和治疗组的用药可显著改善骨性关节炎家兔全血黏度、全血还原黏度、血浆黏度及红细胞聚集指数（P〈0.05或P〈0.01）。结论：从关节软骨及滑膜的光镜观察可以判断,兔膝骨性关节炎模型建立成功;桑生除痹合剂能明显改善血液流变学状态,改善局部血液循环,延缓软骨退化。

Effect of Sangsheng Chubi Mixture on histology and hemorheological index of rabbits with osteoarthritis

Objective： To evaluate the effect of Sangsheng Chubi Mixture on histology and hemorheological index in rabbits with osteoarthritis.Methods： 36 rabbits were randomly divided into 6 groups（each group only 6）,normal group,model group,control group and 3 treatment groups with Sangsheng Chubi Mixture high dose group（treatment groupⅠ）,middle dose group（treatment groupⅡ） and low dose group（treatment group Ⅲ）].The knee osteoarthritis model was replicated by modified Hulth＇s modeling method.Treatment group Ⅰ,treatment groupⅡ,treatment group Ⅲ were given high dose,middle dose,low dose of Sangsheng Chubi Mixture respectively,and control group was given Bones Joints Pill.All rabbits were killed after 12 weeks,the histology change of the articular cartilage and synovial membrane were observed under light microscope,and the hemorheological indexs of 6 groups were observed.Results： Degeneration was found in articular cartilage and synovial membrane under light microscope,whole blood viscosity,whole blood reduced viscosity（WBRV）,plasma viscosity and RBC aggregation index were changed observably,there were highly statistically significant in control group and treatment groups.Conclusion： Establishment of knee osteoarthritis model is successful according to articular cartilage and synovial membrane.The hemorheology and blood circulation are improved and the degeneration of articular cartilage is delayed by Sangsheng Chubi Mixture.
