| 摘 要： | 目的:探讨中医活血健脾补肾法对骨髓间充质干细胞(MSC)移植在结肠炎大鼠肠道定植的影响。方法:雌性SD大鼠以5%2、4、6,三硝基苯磺酸(TNBs)诱导形成结肠炎模型,造模后随机分为模型对照组、MSC移植组、活血组、健脾组、补肾组,第2天将体外分离培养的雄性SD大鼠MSC经尾静脉注射到各组雌性大鼠体内。疗程结束后取各组大鼠结肠组织,以Y染色体的性别决定区段sry作为标志,以确定MSC的肠道定位情况;免疫组化法检测其肠道干细胞标记物Musashi-1的表达。结果:经PCR扩增后,凝胶电泳可以检测到结肠组织雄性性别特异性基因区段sry,MSC移植组阳性率为20%,活血组阳性率为35%,健脾和补肾组阳性率为25%。免疫组化检测到MSC移植组及中药治疗各组大鼠结肠黏膜组织Musashi-1均有阳性表达,其中活血组与MSC移植组比较差异有显著性意义。结论:活血法代表方药桃红四物汤较明显促进MSC在结肠炎大鼠肠道定植。
