摘    要:目的 选择最优的西洋参人参皂苷的提取方法。方法 采用甲醇超声提取法和正丁醇回流加热提取法提取西洋参根中的人参皂苷,利用高效液相色谱法(HPLC)测定人参皂苷Rg1、Re、Rb1、Rc、Rd 含量差异,比较两种提取方法的优劣。结果 两种提取方法提取的人参皂苷Rg1、Re、Rb1、Rc、Rd在测定范围内具有良好的线性关系。甲醇超声提取的Rg1、Re 平均含量均高于正丁醇加热回流提取法(P<0.05),Rb1平均含量低于正丁醇加热回流提取法(P<0.05),Rc、Rd 平均含量与正丁醇加热回流提取法无显著差异(P>0.05)。方法学考察结果显示人参皂苷Rg1、Re、Rb1、Rc、Rd日内精密度RSD良好、平均加样回收率良好。结论 甲醇超声提取的Rg1、Re含量显著高于正丁醇提取,正丁醇加热回流提取法Rb1显著高于甲醇超声提取。依据不同的人参皂苷成分选择最佳的提取方法。

Abstract:Objective To select the best extraction method of ginsenosides in Panaxquinquefolium L. Methods Methanol ultrasonic method and N-butanol reflux method were used to optimize the best extraction method of Ginsenosides. High Performance Liquid Chromatography (HPLC) was established to determine the content of ginsenosides. Results The average content of Rg1 and Re extracted by ultrasonication of methanol is significantly higher than that of n-butanol reflux extraction(P<0.05); while the average content of Rb1 extracted by ultrasonication of methanol is significantly lower extracted by n-butanol reflux extraction(P<0.05).There is no significant difference between the average content of Rc and Rd within the two methods. The content of Rg1 extracted by methanol is higher than that of n-butanol extraction. Methodological investigation showed the best that the inter-day precision and the average recovery rate of Rg1, Re, Rb1, Rc and Rd. Conclusion Rg1 and Re extracted by methanol ultrasonication are significant higher than n-butanol reflux extraction. While, Rb1 extracted by methanol ultrasonicationis significant lower than n-butanol reflux extraction. Thus, the optimal method for ginsenosides extraction of American ginseng should be chosen by different ginsenosides component.
