摘    要: 目的 建立高效毛细管电泳法测定血浆中布比卡因的浓度。方法 取0.5 ml血浆,用乙醚提取后吹干,用1 mmol·L-1 HCl溶解,采用毛细管电泳法进行测定。电泳条件为:分离用缓冲液为20 mmol·L-1 NaH2PO4溶液(Ph4.8),实验温度:25℃,分离电压:30 Kv,紫外检测波长:200 nm,进样:电动进样。结果 本法在0.1~1.0 μg·ml-1范围内呈良好的线性关系和重现性,方法的检测限为0.03 μg·ml-1。结论 本法简便、快速、灵敏,为血浆中布比卡因的测定提供了新的更为理想的方法学手段。

Abstract:OBJECTIVE To establish a high-performance capillary electrophoresis method for the determination of bupivacaine in plasma. METHODS 0.5 ml of plasma was extracted by ethyl ether and then evaporated to dryness.The residue was reconstituted in 1 mmol·L-1of HCl for the determinatin by capillary electrophoresis.The electrophoresis conditions were as following:buffer:20 mmol·L-1of NaH2PO4(pH4.8),temperature:25℃,voltage:30 kV,UV detection at 200 nm ,sampling:the sample for determination was injected antomatically. RESULTS The established method had good linearity and precision within the range of 0.1~1.0 μg·ml-1of bupivacaine.The detection limit was 0.02 μg·ml-1. CONCLUSION This assay was simple,rapid and sensitive.It is a new and ideal analytical means for the determination of bupivacaine in plasma.
