弓形虫感染家兔精液中虫体含量的动态变化

摘    要:目的检测弓形虫感染家兔精液中虫体DNA,观察虫体在精液中动态变化情况。方法用RH株弓形虫速殖子105个/只的剂量腹腔感染雄性家兔16只。在家兔感染前后分别采集精液,-40℃冻存备用,采用两种方法提取精液中总DNA,用实时定量PCR(quantitativereal-timePCR,QRT-PCR)检测弓形虫感染家兔精液中弓形虫DNA,依据对照计算虫体拷贝量,绘制感染后时间与精液中虫体含量的动态曲线图。结果16只雄兔感染前其精液中均不能检测到弓形虫DNA,精液中虫体含量在感染后7wk左右达到高峰期,然后虫体含量逐渐下降。在感染9wk~15wk维持较低水平。结论弓形虫感染雄兔精液中虫体含量随感染后时间不同而变化。

Abstract:The dynamic change on densities of Toxoplasma gondii in semen from infected rabbits was investigated in the present study, in which 16 male rabbits were infected with T.gondii by intra-peritoneal inoculation with 10 5 tachyzoites of the RH strain,and the semen samples collected before and after infection were stored at -40℃ until used for examination. Quantitative real-time PCR assay was used to detect DNA of T.gondii in semen extracted by two different methods, and the density of T.gondii was calculated in reference to the copy numbers ofknown T.gondii DNA. Thereafter, the dynamic figure on density of T.gondii in semen of the infected rabbits was illustrated with Microsoft Excel. It was found that the T.gondii DNA in semen of the infected rabbits could be detected around 7 days after infection and its density gradually rose with a peak value of density about 7 weeks and a gradual decrease afterwards. During 9-15 weeks after infection,it still remained at a low level. It is evidnt that the density of T.gondii in semn of rabbits fluctuates with times of infection.
