ZD1839对A549细胞EGFR及其下游信号分子表达的影响

摘    要: 目的 研究ZD1839对A549细胞EGFR基因及其下游信号通路的影响。方法 利用MTT法检测ZD1839对A549细胞的生长抑制作用,Real-Time PCR检测EGFR及其下游信号分子表达的影响。结果 ZD1839可显著抑制A549细胞的生长,IC50在10 μmol/L左右。加入ZD1839后A549细胞EGFR基因表达为不加ZD1839的1.10倍,Ras基因为1.09倍,MAPK为0.521倍,PI3K为0.164倍,Akt为0.253倍。ZD1839对A549细胞的EGFR、Ras基因表达无影响,使MAPK、PI3K、Akt表达下调。结论 ZD1839抑制EGFR酪氨酸激酶活性,通过影响其下游PI3K/Akt、MAPK信号通路抑制A549细胞生长。

Abstract:Objective Discuss the change of the EGFR expression and downstream signal pathway in A549 cell treated with ZD1839. Methods The inhibition of the A549 cell treated with ZD1839 was mea-sured by MTT assay and Real-Time PCR was used to evaluate the expression of EGFR and downstream signal pathway. Results A549 cell was inhibited by ZD1839 in vitro. After treated with ZD1839, the expression of EGFR was 1.10 fold compared with the cell without ZD1839 and Ras gene was 1.09 fold. The expression of MAPK was 52.1 % of the cell without ZD1839, PI3K 16.4 % and Akt 25.3 %. ZD1839 didn't affect the ex-pression of EGFR and Ras in A549 cell but down regulated the expression of MAPK, PI3k and Akt. Conclu-sion ZD1839 inhibit A549 cell by inhibiting the tyrosine kinase activity of EGFR and the PI3K/Akt, MAPK signal pathway.
