核因子-κB对糖尿病大鼠肾组织P-选择素表达的影响

目的研究糖尿病大鼠肾组织核因子-κB（NF-κB）和P-选择素（P-selectin）的表达，以及NF-κB抑制剂吡咯烷二硫基甲酸酯（PDTC）对其表达的影响。方法30只雄性Wistar大鼠随机分为三组：正常对照组（NC组）、糖尿病组（DM组）、吡咯烷二硫基甲酸酯干预组（DP组），每组10只。喂养8周末检测血糖、糖化血红蛋白、肾重指数及尿白蛋白排泄率。采用免疫组织化学染色技术检测肾组织P—selectin及NF-κB表达。结果8周末，DM组大鼠肾组织中NF-κB和P—selectin表达明显高于NC组（P〈0．01），DP组的表达显著低于DM组，高于NC组（P〈0．01）；P—selectin表达与NF-κB呈正相关（r=0．954，P〈0．01）。结论NF-κB及P—selectin表达在糖尿病大鼠肾组织中明显增加，抑制NF-κB表达可减少P—selectin的表达。

Effects of NF-Kappa B on the Expression of P-selectin in the Kidney of Diabetic Rats

Objective To investigate the expressions of P -selectin and nuclear factor -kappa B （ NF -κB） in the kidney of diabetic rats, and their changes when the expression of nuclear fac- tor - kappa B （ NF-κB） was inhibited by PDTC. Methods 30 male Wistar rats were divided randomly into 3 groups： normal control group （NC group）,diabetic model group （DM group） and diabetic rats treated with PDTC （ a inhibitor of NF-κB） （DP group） with 10 rats in each group. Eight weeks later, blood glucose, glycosylated hemoglobin, kidney weight index and u- rine albumin excretion rate were determined. At the same time, the expressions of P - selectin and NF-κB were detected by immunohistochemistry. Results Expressions of P - selectin and NF-κB in the DM group were significantly higher than those in NC group（P 〈0. 0l ）,the expressions of those in the DP group were significantly lower than those in the DM group, but significantly higher than those in NC group （P 〈 0.01 ）. And the expression of p - selectin showed a strong positive correlation with that of NF-κB （ r = 0. 954, P 〈 0.0l ）. Conclusion In the renal tissue of diabetic rats, the expressions of NF -κB and P - selectin significantly in- crease, and inhibiting the expression of NF-κB can decrease the expression of P -selectin.
