摘    要:目的 应用MTT比色法研究抗小鼠宫颈癌单克隆抗体(McAb)AU14-1介导淋巴因子激活的杀伤细胞Gym-phokine activated killercells,LAK)对靶肿瘤细胞的体外细胞毒作用。方法 通过检测不同数量小鼠宫颈癌U14细胞与加入四甲基偶氮唑盐{[3-(4,5-dimethylthiazof-2-y1)-2,5-diphenyl terazolium bromide],MTT}后形成甲质颗粒所产生的不同吸光度(A)值,建立测定的线性关系。进而应用MTT比色法研究McAb AU14-1介导LAK细胞的体外细胞毒作用。结果 当U14细胞在10~10^5个/孔以内时,活细胞数与A值呈直线相关;LAK细胞对于经AU14-1处理的肿瘤细胞的杀伤率显著高于未经AU14-1处理的肿瘤细胞(P<0.01)。结论 McAb AU14-1可加强LAK细胞对靶细胞的溶解作用。提示特异性宫颈癌单克隆抗体AU14-1介导的细胞毒在治疗宫颈癌中可能是一种有潜在价值的方法。

Abstract:Objective To investigate the cytotoxicity of McAb A U 14-1 mediated LAK against target cervical carcinoma by MTT colorimetric assay.Methods MTT assay was employed to study the relationship betw een A value and different numbers of viable U 14 cells with the aim to esta blish the standard linearity with which the cytotoxic activities of McAb AU 1 4-1 mediated LAK in vitro were evaluated.Results It revealed a l inearity between the different numbers of the cells and A value; The cytotoxicit y mediated by LAK to U 14 cells treated with McAb AU 14-1 was signifi cantly higher than those not treated with it.Conclusion McAb AU 14-1 could enhance the cytotoxicity of LAK to U 14 cells,suggesting its ability to direct ADCC make this antibody promising candidate for cervical cance r therapy.
