端粒酶抑制剂对顺铂不同敏感性卵巢癌细胞株的影响

摘    要:目的:通过检测顺铂(CDDP)敏感株和耐药株在经CDDP处理前后的端粒酶活性变化,了解端粒酶活性与CDDP耐药性的关系,及端粒酶抑制剂叠氮胸苷(AZT)和全反式维甲酸(AT-RA)对CDDP不同敏感性的卵巢癌细胞株的影响,试图寻找一个解决常规化疗耐药问题的新方法。方法:将卵巢癌CDDP敏感株A2780和耐药株CAOV3、OVCAR3在不同浓度的CDDP、AZT和ATRA中培养,TRAP-ELISA法检测端粒酶活性,MTT法检测药物对细胞增殖的影响。结果:未加CDDP时,敏感株和耐药株的端粒酶活性没有明显差别,经CDDP处理后敏感株的端粒酶活性明显降低,而耐药株没有明显变化。端粒酶抑制剂对CDDP敏感株和耐药株的细胞增殖都有明显的抑制作用。结论:通过不同方式抑制端粒酶活性,均可抑制CDDP敏感株和耐药株的细胞增殖。端粒酶的活性与卵巢癌细胞株对CDDP的耐药性有关。

Abstract:Objective:In order to understand the relation between telomerase activation in ovarian cancer cell line and CDDP resistance by detecting telomerase activation in CDDP-sensitivity and CDDP-resistant strains treated by CDDP.And to solve the problem of resistance to routine chemotherapy by studying the effect of telomerase inhibitors AZT and ATRA to ovarian cancer strains with different sensitivity to CDDP.Methods: After incubated with different concentration of CDDP,AZT and ATRA,telomerase activation and cell proliferation of the CDDP-sensitivity strain A2780 and the CDDP-resistant strains CAOV_3 and OVCAR_3 were detected with TRAP-ELISA and MTT,separately.Results: There was no difference in telomerase activation between CDDP-sensitivity and CDDP-resistance strain before incubated with CDDP.After treaded by CDDP,the telomerase activation of the CDDP-sensitivity strain decreased significantly while the CDDP-resistant strains had no evidently variant.The cell proliferation was markedly inhibited by Telomerase inhibitors in both kinds of cell strains.Conclusions: Through inhibiting telomerase activation in different way,cell proliferation of both sensitivity and resistant strains to CDDP can be inhibited.Telomerase activation was related to CDDP-resistance of ovarian cancer cell line.
