罗格列酮对高糖时人肾小球系膜细胞单核细胞趋化蛋白-1表达的影响

摘    要:【目的】探讨高糖对人肾小球系膜细胞(HRMC)单核细胞趋化蛋白-1(MCP-1)表达的影响,以及过氧化物酶增殖体激动剂罗格列酮对其影响及作用机制。【方法】将HRMC分为三组处理:①正常对照组(糖浓度5.5mmol/L);②高糖(30mmol/L)组;③高糖加罗格列酮(5μmol/L,10μmol/L)组。用反转录-聚合酶链式反应(RT-PCR)方法检测MCP-1mRNA表达,用EIISA方法测定细胞上清MCP-1蛋白浓度。【结果】高糖刺激系膜细胞后MCP-1mRNA表达明显升高,蛋白浓度增加,加入罗格列酮后MCP-1表达明显下降。且随罗格列酮浓度增大,作用更明显。【结论】高糖刺激HRMC,其MCP-1表达增强,可能是糖尿病肾病(DN)发病机制中的一个重要途径。罗格列酮能抑制MCp-1mRNA表达及蛋白合成,说明其具有抑制炎症的作用,在预防DN的发生发展中可能有重要意义。

Abstract:[Obiective]To study the effect of high glucose on MCP-1 production from cultured human mesangial cells and the protective effect and mechanism of rosiglitazone. [Methods]Human mesangial cells were used as target cells. MCP-1 expression in supernatants was determined by ELISA and MCP-lmRNA in human mesangial cells was detected by RT-PCR. High glucose increased the expressing of MCP-lmRNA and MCP-1 protein concentration at 4 hours, 24 hours or 48 hours. [Results]Rosiglitazone could inhibit these effects of high glucose. [Conclusion]The expression of MCP-1 in human mesangial cells can be induced by high glucose and rosiglitazone can inhibit these effects.
