摘    要:目的建立高效液相色谱法测定塞来昔布中三氟乙酸的残留含量。方法色谱柱:采用HypersilODS柱(4.6mm×250mm,5㈣,检测波长210nm,流动相:0.08%磷酸溶液㈣3.0)-甲醇(95:5),流速1.0ml/min,柱温:35℃。结果三氟乙酸浓度在2.05~12.12μml范围内,线性关系良好,r=0.9998;检测限为1.0ng;加样回收率平均为98.57%,RSD为0.99%。结论此法可用于塞来昔布中三氟乙酸含量的测定,准确度高。

Determination of trifluoroacetic acid in celecoxib by HPLC

Abstract:Objective To establish a HPLC method for determination of the content of trifluoroacetic acid in celecoxib. Methods Hypersil ODS column(4.6 mm × 250 mm, 5 μm) and 0.08% phosphoric acid so -lution( pH 3.0)-methanol(95:5) as the mobile phase at a flow rate of 1.0 ml/min were used with a column temperature of 35 ℃ and detection wavelength of 210 nm. Results The standardcurve of trifluoroacetic acid was linear in the range of 2.05-12.12μg/ml, r=- 0.9998.The detection limit was 1.0 ng. The average recovery rate of method was 98.57%, and RSD was 0.99%. Conclusion The method of content determination is practicable and accurate. It can be used for content determination of trifluoroacetic acid in celecoxib.
