摘    要:目的:建立巴戟天组织培养再生体系,诱导培育出巴戟天多倍体.方法:以巴戟天的幼胚为外植体诱导出愈伤组织,再用不同浓度的秋水仙素处理愈伤组织,流式细胞仪检测染色体数.结果:以秋水仙素质量浓度为500 mg·L-1处理5d效果最好,多倍体诱导率可达18.4%,诱导培育出的巴戟天多倍体根肥大增粗.结论:以巴戟天的幼胚为外植体具有消毒容易、诱导率高、分化程度低的优点;诱导出的多倍体具有一定的推广应用价值.

Abstract:Objective : To establish an effective protocol for plant generation and induce polyploidy of Morinda officinalis. Method : Callus was induced from immature embryo of M. officinalis and polyploidy was inducted by using colchicine treatment method. Chromosome was detected by flow cytometry. Result and Conclusion : The highest induction rate of polyploidy was 18.40%, which was obtained with 500 mg·L-1 colchicine treatment for 5 days. Roots of polyploid were bigger than diploid. Advantages of using immature embryo as explants are easy for sterilization, higher rate of callus induction and low degree dedifferentiation. The induced polyploidy of M. officinalis may have a value for spread of cultivation.
