摘    要:目的探讨奎硫平对血糖影响的可能机制。方法正常SD大鼠20只,随机分为对照组和奎硫平组,分别于给药前、给药1、2、3、4周后测体重及空腹血糖;给药4周后放免法测血胰岛素、C肽及RT-qPCR法分析胰腺组织GLUT2mR-NA的表达。结果 (1)给药2、3、4周后,奎硫平组大鼠体重均大于对照组(P<0.05);每周体重增加量奎硫平组均大于对照组,但只有第1周差异有显著性(P<0.01)。(2)奎硫平组空腹血糖呈增长趋势,给药2、3、4周后均高于给药前(P<0.05)。(3)奎硫平组大鼠血胰岛素、C肽水平高于对照组,差异无显著性(P>0.05)。(4)奎硫平组大鼠GLUT2mRNA表达水平低于对照组(P<0.01)。结论奎硫平可以降低GLUT2mRNA的表达,这可能是长期应用奎硫平后引起血糖异常的发生机制之一。

The effects and possible mechanism of quetiapine on glucose regulation of rats
Abstract:Aim To discuss the possible mechanism of quetiapine's effects on glucose regulation.Methods Twenty SD rats were randomly assigned into control group and quetiapine group.Weight and fasting glucose(FBG) were measured before administration,as well as 1,2,3 and 4 weeks after drug administration;4 weeks after drug administration,blood insulin and C-peptide were tested by radioimmunoassay administration;at the same time,pancreatic GLUT2 mRNA expression level was detected by RT-qPCR.Results(1) on week 2,3,and 4,the rats in quetiapine group weighted more than control group(P<0.05);and weekly weight gain of quetiapine group was higher than that of control group,but the difference was only significant in the first week(P<0.01).(2) The increasing trend of FBG was observed in quetiapine group;2,3 and 4 weeks after drug administration,the FBG was higher than pretreatment(P<0.05).(3) Both the level of insulin and that of C-peptide of quetiapine group were higher than those of control group,but there was no statistically significant difference(P>0.05).(4) The expression level of GLUT2 mRNA of quetiapine group was lower than that of control group(P<0.01).Conclusion Quetiapine could decrease the expression level of GLUT2mRNA,which may be one of the causes of glucose abnormality followed by chronic administration of quetiapine.
