胃癌化疗体外药敏试验研究(附88例分析)

目的检测目前常用化疗药物对体外培养的原代胃癌细胞的敏感性.方法用改良MTT法对88例胃癌患者的新鲜标本进行体外肿瘤细胞原代培养和化疗药物敏感性检测,并对其结果进行比较.结果试验成功率为94.3%,以细胞抑制率30%为判断标准的14种化疗药物中,5-FU、MMC、PYM、VP-16和DDP具有较高的敏感性（P＜0.05）.以上5种药物之间比较,其敏感性差异无显著意义（P＞0.05）;肿瘤分化程度的高低与肿瘤药敏无明显关系.结论体外胃癌细胞原代培养及化疗药物敏感试验对指导临床医生选择敏感化疗药物开展个体化化疗具有指导作用.

Study on sensitivity of human gastric carcinoma to chemotherapeutic drugs in vitro (report of 88 cases)

Objective To investigate the sensitivity of gastric carcinoma to different chemotherapeutic drugs by the method of primary culture of gastric carcinoma cells.Methods The gastric carcinoma cells taken from fresh samples of 88 patients were cultured in vitro to test the sensitivity of these cells to chemotherapeutic drugs using modified MTT method.Results The test success rate was 94.3%. 5-FU, MMC, PYM, VP-16 and DDP were effective to gastric cells among 14 drugs judged by the criterion of 30% inhibitory rate. No effects were observed in the other 5 drugs judged by the criterion of 50% inhibitory rate. there were no significant differences among these sensitive drugs above, and the degrees of differentiation of gastric carcinoma were not related to the drug sensitivity(P>0.05).Conclusion Drug sensitivity test for gastric carcinoma is feasible, this may gave some guidance to physicians for selecting the sensitive drugs.
