山药多糖对正常小鼠与实邪证模型小鼠免疫功能的影响

摘    要:目的:研究山药多糖对正常小鼠与实邪证模型小鼠免疫功能的影响。方法:40只ICR小鼠随机均分为正常对照(等容生理盐水)组与山药多糖高、中、低剂量(400、200、100 mg/kg)组;40只实邪证模型小鼠随机均分为实邪证模型(等容生理盐水)组与山药多糖高、中、低剂量(400、200、100 mg/kg)组,另以10只ICR小鼠为正常对照(等容生理盐水)组,均灌胃给药,每天1次,连续4周。进行碳粒廓清实验,绵羊红细胞诱导小鼠迟发型变态反应,测定小鼠血清白细胞介素(IL)-2含量。结果:与正常对照组比较,山药多糖高、中剂量组小鼠廊清指数(K)、吞噬指数(α)、足肿胀程度、血清IL-2含量增加,差异有统计学意义(P<0.05);与实邪证模型组比较,山药多糖高剂量组小鼠K、α、足肿胀程度、血清IL-2含量减少,差异有统计学意义(P<0.05)。结论:山药多糖对正常小鼠免疫力有促进作用,但对实邪证模型小鼠免疫力则有削弱作用。因此,临床用药时应注意具体情况,以免起到相反效果。

Effects of Dioscorea opposita Polysaccharides on Immune Function in Normal Mice and Mice with Dampness and Blood-stasis Syndrome

Abstract:OBJECTIVE: To study the influence ofDioscorea opposita polysaccharide on immune function in normal mice and mice with dampness and blood-stasis syndrome. METHODS: ICR mice were randomized into normal control group (constant volume of normal saline), D. opposita polysaccharide high-dose, medium-dose and low-dose groups (400, 200, 100 mg/kg) ; 40 mice with dampness and blood-stasis syndrome were randomized into model group (constant volume of normal saline),D, opposita polysaccharide high-dose, medium-dose and low-dose groups (400, 200, 100 mg/kg) ; other ICR mice were included in normal control group, they were given relevant medicines intragastrically once a day for consecutive 4 weeks. Carbon clearance test was conducted, and sheep red blood cell induced delayed hypersensitivity reaction in mice. The serum level of IL-2 in mice was determined. RESULTS: Compared with normal control group, clearance index (K), phagocytic count (a), mice foot swelling degree and the content of serum IL-2 increased in D. opposita polysaccharide high-dose and medium-dose groups ; there was statistical sig- nificance(P〈0.05), Compared with model group, K, the value of α, mice foot swelling degree, the serum level of IL-2 decreased in D. opposita polysaccharide high-dose group; there was statistical significance (P〈0.05). CONCLUSIONS: D. opposita polysaccharide promotes immune function in normal mice but weaken immune function of mice with dampness and blood-stasis syndrome. Therefore, we should pay attention to the specific circumstances of patients in clinical medication, so as to avoid the opposite effect.
