牛奶酪蛋白水解肽血脑屏障透过性的研究

摘    要:目的研究牛奶酪蛋白水解肽的血脑屏障透过性。方法利用一种大鼠静脉给药后抽取脑脊液的动物模型,静脉注射牛奶酪蛋白水解肽后,在不同的时间点分别取血清样品和大鼠脑脊液样品,样品经过预处理后,用高效液相色谱(HPLC)进行分析。结果结果显示该多肽混合物在体循环中极易被代谢,在静脉给药6 min之后就无法在血清中检测到任何可见的样品峰。而在6 min之前不同时间点的血清检测结果中,样品峰强度随着时间的递增逐渐降低。同时在脑脊液样品的检测中,结果显示在3 min和4 min这两个时间点的脑脊液样品中出现了一个保留时间相同的新峰。其保留时间与其中一个多肽样品峰非常的接近。结论牛奶酪蛋白水解肽中的某种成分通过血脑屏障进入脑内,也可能是脑脊液中的某些分子的浓度发生了变化。该结果同时提示我们食源性的天然多肽分子有可能能够直接通过血脑屏障进入中枢伸进系统CNS,从而发挥相应的神经生物学功能。

Abstract:Objective To study the blood-brain barrier perviousness of the hydrolyzed peptides of casein which derived from milk.Methods A type of hychrolyzed peptide was injected into rat carotid vein and detected its concentration in serum and cerebrospinal fluid(CSF) at different time spot.The peptide sample was analyzed by HPLC after pretreatment.Results The sample peptides were very easy to be metabolized in systemic circulation and they were undetectable after intravenous administration for 6 minutes,and the peak intensity of the peptides decreased as time goes by within 6 minutes.Meanwhile two new peaks appeared in the CSF sample at 3 and 4 minutes spot.The retention time of those new peaks were very close to one of the peaks of peptide sample.Conclusion This result suggest that some peptides in the bovine hydrolyzed peptides of casein could cross the blood-brain barrier or the concentration of some molecules in the CSF have been changed.Meanwhile food-origin natural peptides may be possible to cross the BBB and pass into the CNS to change the function or even the structure of central nervous system.
