变应性鼻炎与肿瘤坏死因子水平及肿瘤坏死因子-863C/A基因多态性的关系初步探讨

摘    要:目的探讨肿瘤坏死因子水平及肿瘤坏死因子.863C/A基因多态性与变应性鼻炎的关系。方法用放免法测定TNF水平,运用多聚酶链反应技术检测90名无亲缘关系的尘螨变应性鼻炎患者和107名无血缘关系的健康汉族人肿瘤坏死因子.863C/A基因多态性。结果变应性鼻炎组肿瘤坏死因子TNFa-863C/C、A/C、A/A表型频率分别为:0.7222、0.2667、0.0111,对照组分别为0.7477、0.2429、0.0094;变应性鼻炎组TNFa-863A、TNFa.863C基因频率分别为0.8556、0.1444,对照组分别为O.8692、0.1308;TNFa.863各种基因型频率在变应性鼻炎组与正常对照组之间差异无统计学意义(P〈005)。而血浆中TNF水平,变应性鼻炎组明显高于对照组,两组之间比较差异有统计学意义(P〈0.05)。结论血清TNF水平显著升高,提示炎性反应在变应性鼻炎病程中有重要作用,肿瘤坏死因子TNFa-863C/A基因多态性与变应性鼻炎无明显相关。

Abstract:Objective To investigate the relationship between plasma levels of tumor necrosis factor(TNF) or its polymor- phism and allergic rhinitis. Methods Plasma level of TNF was measured by RIA the polymorphism of TNFα-863 C/A gene was screened by PCR-RELP. Results The gene frequency of TNFct-863 C/A in allergic rhinitis group was as followed ; TNFa-863 C/C, A/C, A/A genotype frequency: 0.7222, 0.2667, 0.0111,TNFct-863 C, NFα-863 A gene frequency: 0.8556, 0.1444. TNFct- 863 C/C, A/C, A/A genotype frequency in control group was as followed: 0.7477, 0.2429, 0.0094,TNFcx-863 C, TNFα-863 A gene frequency: 0.8692, 0.1308, there were no significant differences in the frequencies of TNFα-863 gene between allergic rhinitis patients and control group (P〉0.05). allergic rhinitis group showed significantly higher plasma level of TNF than control group (P〈0.05). Conclusion Plasma TNF increased in allergic rhinitis, the results suggested: TNF play important role in the process of allergic rhinitis and no evident association was found between TNFα-850C/T polymorphism and allergic rhinitis.
