四种不同组织来源LAK细胞的体外扩增及抗肿瘤作用的比较

摘    要:目的:比较四种不同组织来源LAK细胞的体外扩增及抗肿瘤作用。方法:选用脐带血制备LAK细胞,并与临床常用的外周血、胎脾及胸腺LAK细胞在体外扩增及抗肿瘤作用方面比较。结果:脐带血的扩增能力显著高于其余3种LAK细胞(P<0.01),每份脐带血LAK细胞总数可增殖至2.5×1010;抗肿瘤活性以脐带血LAK对SMMC-7221肝癌细胞出现最早(第3d)、活性最强(85%±3%),明显强于其余三者(P<o.01);对K562的杀伤作用仍属脐带血LAK最强;而胸腺LAK的活性最弱。结论:脐带血LAK细胞体外增殖快,杀伤活性强,可用于肿瘤的过继性免疫治疗。

Comparison of expansion and cytotoxicity in vitro of lymphokine-activated killer cells from four different tissues

Abstract:Objective: To compare the expansion and cytotoxicity in vitro of lymphokine-activated killer (LAK) cells from four different tissues. Metheds: This study prepared LAK cells from umbilical cord bllld and compared them with common LAK cells from donor,fetal spleen and thymus on expansion and cytotoxicity. Results:The expansion of LAK cells from umbilical cord blood within two weeks in vitro was maximal among four kinds of LAK cells (P<0. 01 ). The maximal cells numbers of each umbilical cord blood were 2. 5×1010,whichaccorded with international standard. The cytotoxicity of umbilical cord blood against SMMC-7221 in vitro occured earliest (on the third day) and showed maximal lysis rate (85±3% ),the most powerful among the all LAK cells (P<0. 01 ). Also these LAK had the highest cytotoxicity against K562 (71±2 % ). But the activity of thymus LAK cells against two target cells was the lowest. Conclusion :These results suggest that umbilical cord blood LAK cells have the advantages of rich sources,simple preparation,maximal expansion,and powerful cyto-toxicity. They will be a new high qualitative source of LAK cells in clinical trials.
