汽油对皮肤细胞DNA、蛋白质及皮脂合成能力的影响

摘    要:目的 研究溶剂汽油对皮肤细胞的损害作用机制。方法 采用皮肤角蛋白细胞和成纤维细胞原代培养技术,以及^H-胸腺嘧啶核苷(^3H-TdR)、^3H-脯氨酸(^3H-Pro)、^3H-亮氨酸(^3H-Leu)和^14C-亚油酸掺入实验方法,观测溶剂汽油对皮肤细胞DNA、蛋白质及皮脂合成能力的影响。结果 接触0.01%的溶剂汽油后,角蛋白细胞的^3H-TdR和^3H-Pro的掺入受到明显抑制,抑制率分别为68.5%、45.1%和40.6%;而在接触0.001%溶剂汽油时,角蛋白细胞的^3H-Leu和^14C-亚油酸的掺入就已表现出显著性降低,抑制率分别为20.2%和41.2%。皮肤细胞的掺入抑制率与接触溶剂汽油的剂量有关。结论 溶剂汽油对皮肤细胞具有一定的毒效应,可干扰皮肤细胞的正常生理代谢过程,影响其DNA、蛋白质及脂质合成能力,使皮肤各项重量功能受到影响,这可能是溶剂汽油造成皮肤损害的机制之一。与成纤维细胞相比,角蛋白细胞更易受到溶剂汽油的影响。

Abstract:Objective To study the cellular and molecular mechanism of gasoline induced adverse effects on skin, particularly on keratinocyte and fibroblast in vitro Methods The primary cell culture of keratinocyte and fibroblast were treated with 0, 0 001%, 0 01%, 0 1% and 1 0% gasoline, respectively 3?H thymidine ( 3H TdR), 3?H leucine ( 3H Leu), 3?H proline ( 3H Pro) and 14 ?C linoleic acid incorporation tests were applied to elucidate their capacity of synthesizing DNA, protein and sebum Results The incorporation of 3?H TdR in keratinocyte and 3?H TdR and 3?H Pro in fibroblast inhibited significantly after exposure to 0 01% gasoline ( P <0 05), with inhibition rates 68 5%, 45 1% and 40 6% for 3?H TdR in keratinocyte, and 3?H TdR and 3?H Pro in fibroblast, respectively Significant depression in incorporation of 3?H Leu and 14 ?C linoleic acid in keratinocyte were found even in the group treated with 0 001% gasoline ( P <0 05), with inhibition rates of 20 2% and 41 2%, respectively Conclusions Solvent gasoline has certain toxic effect on keratinocyte and fibroblast, intervening their normal metabolic and physiological process and affecting their ability of synthesizing DNA, protein and sebum, and their physiological functions, which could be one of the mechanisms causing skin damage by gasoline The results also indicated that keratinocyte was more susceptible to gasoline than fibroblast
