铁离子对副百日咳鲍特菌生长的影响

摘    要:目的研究铁离子对副百日咳鲍特菌体外培养的影响。方法在ss肉汤培养基中分别加入不同浓度的铁离子螯合剂,并设加硫酸亚铁的铁离子组为对照组,绘制不同培养条件下副百日咳鲍特菌的生长曲线,比较副百日咳鲍特菌在低铁和高铁环境下的生长情况。采用721可见分光光度计测定600nm波长下的菌液吸光度值,副百日咳鲍特菌的生长起始吸光度值为0.15—0.20,37℃振荡培养。结果对照组的副百13咳鲍特菌在22h开始进入对数生长期,经20h后进入平台期。在加入终浓度为0、50、100、200μmol/L铁离子螯合剂的ss肉汤培养基中,副百日咳鲍特菌均在46h后进入对数生长期,对数生长期的时间与对照组相近。当铁离子螯合剂浓度达300μmol/L时,副百日咳鲍特菌不再生长。结论副百日咳鲍特菌的生长需铁离子的存在,副百日咳鲍特菌的生长在高浓度和低浓度铁离子环境下有显著差异。

Influence of iron ion on the growth of Bordetella parapertussis

Abstract:[ Objective ] To study the influence of iron on the growth of Bordetella parapertussis (B-pp) in vitro. [ Methods ] The iron ion chelating agents with different concentrations were respectively injected into the Stainer-Scholte ( SS } synthetic broth medium, and ferrous sulfate group was regarded as the control group. The growth curves of B-pp under different culture conditions were drew, to compare the growth status of B-pp between low iron and high iron environment. The absorbance value of bacteria liquid was measured at 600 nm by 721-spectrophotometer. The initial absorbance value of B-pp was 0.15-0.20, and the oscillating culture was conducted at 37 ℃. [ Results] The B-pp in the control group reached the exponential phase after 22 hours, and then began to plateau after 20 hours. In the SS synthetic broth medium with iron ion chelating agent of 0, 50 ~mol/L, 100 pLmol/L and 200 ~moL/L, the B-pp reached the exponential phase after 46 hours, which the durations were similar to that of the control group. Furthermore, the growth of B-pp had not been observed in SS medium with iron ion chelating agent of 300 μmol/L. [ Conclusion] The growth of B-pp is closely related to the iron ion, and there is a significant difference in the growth of B-pp between low iron ion con- centration and high iron ion concentration environment.
