血管内皮生长因子C在人乳腺癌细胞株MCF-7及MCF-7/Adr中的表达

摘    要:背景与目的:血管内皮生长因子C(vascular endothelial growth factor C,VEGF-C)是近期发现的血管内皮生长因子(vascular endothelial growth factor,VEGF)家族的新成员。研究发现VEGF-C可特异性作用于淋巴管内皮细胞,并在多种人类肿瘤中表达。为探索VEGF-C在肿瘤发生,发展中的作用,我们分别检测了VEGF-C mRNA蛋白在人乳腺癌细胞株MCF-7及其耐药株MCF-7/Adr中的表达。方法;根据VEGF-C基因序列,设计合成地高辛标记的特异性寡核 探针,运用原位杂交方法检测培养的细胞株MCF-7和MCF-7/Adrk VEGF-C mRNA的表达,并运用免疫组织化学方法检测两种细胞中VEGF-C蛋白的表达,实验中用缓冲液分别代替杂交液和一抗作阴性对照。结果:原位杂交法检测到MCF-7和MCF-7/Adr细胞的胞浆中有阳性蓝色颗粒,免疫组化检测发现两种细胞的胞浆中均有阳性棕黄色颗粒,而阴性对照细胞的胞浆中则均无阳性颗粒。结论:人乳腺癌细胞株MCF-7及其耐药株MCF-7/Adr细胞能够转录VEGF-C mRNA,并在其细胞浆中翻译合成相应的蛋白。

Abstract:Background &Objective:Vascular endothelial growth factor C(VEGF-C)discovered recently is a new member of vascular endothelial grow th factor(VEGF)family.Studies have shown that VEGF-C is one specific regulating factor for lymphatic endothelial cells,and it expresses in a variety of human tumors.In order to explore the r ole of VEGF-C in the process of human tumor formation and development,the authors designed this study to examine the expressions of VEGF-C mRNA and protein in human breast cancer cell line MCF-7and drug resistant cell line MCF-7/Adr.Methods :According to the VEGF-C gene sequence,A specific oligonucleotide probe labeled with digoxin was designed an d constructed;Then in situ hybridiza tion was applied to detect the expression of VEGF-C mRNA in the cultured hu man breast cancer cell lines MCF-7and MCF-7/Adr respectively;Finally immunohistochemistry was performed to examine the expression of VEGF-C protein in both cell lines.Results:Blue positive staining granules for VEGF-C mRNA were observed in the cytoplasm of these cells by in situ hybridization where br own positive staining granules for VEGF-C protein were also observed by immunohistochemistry.While there were no positive staining granules in neith er control cells.Conclusion:Human breast cancer cell lines MCF-7and MCF-7/Adr can transcribe VEGF-C mRNA and translate corresponding protein in their cytoplasm.
