线粒体DNA缺失对人乳腺癌MDA-MB-231细胞药物敏感性的影响

摘    要:目的 探讨线粒体DNA缺失对人乳腺癌细胞药物敏感性的影响。方法 采用溴化乙锭(EB)处理获得人乳腺癌细胞MDA—MB-231的线粒体DNA缺失(rho^0 MDA—MB-231)细胞,电镜下观察细胞形态改变,胶原凝胶包埋三维立体培养法(CD—DST)对比分析细胞的药物敏感性,免疫组织化学方法检测P-糖蛋白(P—gP)、乳腺癌耐药蛋白(BCRP)的表达。结果 与MDA—MB-231细胞比较,rho^0 MDA—MB-231细胞线粒体数量增多,嵴消失呈空泡样改变,对化疗药物相对抵抗,P—gP、BCRP的表达明显增加。结论 线粒体的损伤可能参与了乳腺癌细胞耐药表型的形成。
