摘    要:目的 观察通络醒脑泡腾片对快速老化小鼠亚系8(SAMP8)海马的NeuN蛋白及其超微结构的影响.方法 将SAMP8小鼠随机分为SAMP8组、安理申组、通络醒脑泡腾片组(高、中、低剂量),另取抗快速老化小鼠亚系1(SAMR1)作为对照组,分组后连续给药60d,采用Morris水迷宫和避暗法评价其学习记忆的改变;采用免疫组化法考察海马Aβ和NeuN蛋白的表达;采用电镜法考察海马的超微结构.结果 通络醒脑泡腾片能够缩短SAMP8模型小鼠的潜伏期、明显地增加平台所在象限的时间百分比和进入目标象限的次数,可显著地减少(避暗)错误次数和延长逃避潜伏期;能够下调海马Aβ的表达并上调NeuN蛋白的表达,可显著地减少核糖体的丢失,降低线粒体的肿胀度,改善海马的超微结构.结论 通络醒脑泡腾片可通过抑制Aβ沉积,提高NeuN的表达来修复神经元,保护线粒体,从而提高SAMP8小鼠的学习记忆能力.

Abstract:OBJECTIVE To observe the effect of Tongluo Xingnao effervescent tablet (TLXNET) on the expression of NeuN and ultrastructural in the hippcampus of senescence accelerated mouse/prone 8 (SAMP8).METHODS The SAMP8 mice were randomly divided into five groups and senescence accelerated mouse/resistance 1 (SAMR1) as the blank control group and then treated them 60 days continuously.Theirs learning and memory were evaluated with the step-through task and Morris water maze.The ultrastructural was observed by TEM and the expression of Aβ and NeuN with immunohistochemical in the hippcampus was investigated.RESULTS TLXNET could extend the latency and reduce error times of SAMP8 in the passive avoidance,and also could obviously increase the percentage of target quadrant,times of entering the target quadrant and reduce the latency in the Morris water maze.Furthermore,TLXNET could observably decrease the level of Aβ,enhance the expression of NeuN and protect mitochondria in hippcampus of SAMP8.CONCLUSION TLXNET can ameliorate the pathology variation,renovate neurons,protect the mitochondria and increase the learning and memory in SAMP8.
