摘    要:目的 采用一测多评法同时测定黄连须中5种生物碱(盐酸小檗碱、盐酸药根碱、盐酸表小檗碱、盐酸黄连碱和盐酸巴马汀)的含量.方法 以盐酸小檗碱为内参物,采用HPLC法测定其与盐酸药根碱、盐酸表小檗碱、盐酸黄连碱、盐酸巴马汀的相对校正因子,利用该相对校正因子计算其他4种生物碱的含量;同时利用外标法测定5种生物碱的含量,比较两种测定方法的差异,验证一测多评法的可行性和准确性.结果 在线性范围内,盐酸小檗碱与盐酸药根碱、盐酸表小檗碱、盐酸黄连碱、盐酸巴马汀的相对校正因子分别为1.128、1.008、1.070、1.025,在不同实验条件下,相对校正因子的重复性良好,5种生物碱成分含量的计算值与实测值间无显著性差异.结论 一测多评法同时测定黄连须中5种生物碱的含量是可行的、准确的.

Abstract:OBJECTIVE To establish a Quantitative analysis of multi-components with single marker (QAMS) for the simultaneous determination of five kinds of alkaloids (berberine,jatrorrhizine,epiberberine,coptisine and palmatine) in Coptidis Rootlet.METHODS An HPLC method was established to determine the five alkaloids.Berberine was used as the internal reference,the relative correction factors(RCFs) of the other four alkaloids were set and determined by HPLC.The contents of the other four alkaloids were calculated using their RCFs,which were compared with those determined by External standard method.The feasibility and accuracy of QAMS method were verified.RESULTS Within a certain range,the RCFs of jatrorrhizine,epiberberine,coptisine and palmatine to berberine were 1.128,1.008,1.070 and 1.025,respectively,with the good repeatability in different experimental conditions.There was no significant differences between the QAMS method and External standard method.CONCLUSION The QAMS method is feasible and accurate for the simultaneous determination of five kinds of alkaloids in Coptidis Rootlet.
