参归仁合剂对产后抑郁大鼠雌激素和额叶p-CREB及p-ERK1/2的影响

目的 探讨补虚化淤方药参归仁合剂对产后抑郁大鼠模型雌激素(E2)的水平、额叶环磷腺苷反应元件结合蛋白(CREB)、细胞外调节蛋白激酶(ERK1/2)蛋白磷酸化水平表达的影响.方法 将25只SD大鼠,随机分为正常对照组、模型组、参归仁高、低剂量组(14.0、3.5 g·kg-1)、左洛复(盐酸舍曲林)对照组(4.5 mg·kg-1).采用灌胃给药方式,连续30 d.分别取血、额叶组织,采用放射免疫法检测大鼠的E2水平,采用免疫印迹法检测额叶p-CREB、p-ERK1/2的表达水平.结果 与正常组比较,模型组大鼠血清中E2、p-CREB、p-ERK1/2表达的水平明显降低;与模型组比较,参归仁高、低剂量组和左洛复组的额叶p-CREB的表达量明显升高,参归仁低剂量组和左洛复组的额叶p-ERK1/2表达量升高.结论 参归仁合剂能够明显提高PPD大鼠的E2水平,促进额叶p-CREB、p-ERK1/2的表达上调.

Effects of Shenguiren mixture on estrogen,p-CREB and p-ERK1/2 in the postpartum depression model rats

OBJECTIVE To investigate the effect of Shenguiren mixture (SGRM) on estrogen (E2),phosphor cAMP-response element binding protein(p-CREB)and phosphor extracellular regulated protein kinases (p-ERK) in postpartum depression model rats.METHODS Twenty-five male SD rats were randomly divided into 5 groups:Normal group,model group,high dose SGRM treated group (14.0 g· kg-1),low dose SGRM treated group (3.5 g· kg-1) and Sertraline hydrochloride (4.5 mg· kg-1) group.The rats accepted 30 days intragastric administration,and then blood and frontal lobe of brain were taken.The level of serum estrogen was analysezed by radioimmunoassay.Phosphor CREB,ERK1/2 in frontal lobe of brain detected by Western blot.RESULTS Compared with the normal group,the level of the E2 p-CREB,p-ERK1/2 in the model group were decreased significantly.Compared with the model group,the expression of p-CREB in high,low dose SGRM treated groups and Sertraline hydrochloride group were increased significantly,and so was p-ERK1/2 in low dose SGRM treated group and Sertraline hydrochloride group.CONCLUSION SGRM can obviously improve the level of E2,p-CREB,p-ERK1/2.
