塞北紫堇总生物碱对异丙肾上腺素致心肌缺血损伤的保护作用及机制

摘    要:目的 观察塞北紫堇总生物碱(CITA)对异丙肾上腺素(Iso)致大鼠实验性心肌缺血的保护作用,并探讨其可能的作用机制.方法 将SD大鼠按体重随机分为6组,分别ig 5、20、100 mg·kg-1 CITA,20 mg·kg-1心得安以及等量蒸馏水,每天1次,连续8d,末次给药30 min后,于腹腔注射350 mg·kg-1水合氯醛麻醉,稳定20 min,记录正常心电图,给予各组大鼠皮下多点注射1 mg· kg-Iso,对照组则注射等体积的生理盐水,记录注射后20 min内心电图的T波以及J点的变化.试验结束时,经股动脉取血,测定血清中谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)和丙二醛(MDA)的含量.结果 与对照组比较,在皮下注射Iso 5、20 min时,大鼠心电图的T波明显升高,有统计学意义,而CITA各剂量组均有降低其T波高度的趋势,但作用较弱,与模型组比较,无统计学意义;与对照组比较,心电图J点高度明显降低,有统计学意义,与模型组比较,100 mgCITA可显著对抗Iso降低J点的作用,有统计学意义;与对照组比较,血清中MDA、GSH-Px的含量显著增加,有统计学意义,与对照组比较,其血清中SOD的活性变化不大,无统计学意义.与模型组比较,100、5 mg· kg-CITA可明显降低Iso大鼠血清中MDA、GSH-Px的水平,有统计学意义.结论 CITA对Iso引起的急性心肌缺血损伤具有保护作用,其作用可能与减弱心肌收缩力、扩张冠状动脉血管、减少自由基生成、阻断过氧化脂质反应有关.

Abstract:OBJECTIVE To explore the protective effect of Corydalis impatiens total alkaloids(CITA) on acute myocardial ischemia injury induced by isoproterenol(Iso) in rats and its possible mechanism.METHODS SD rats were randomly divided into six groups according to different administrated dose,ig with 5 mg· kg-1,20 mg· kg-1,100 mg· kg-1 CITA,20 mg· kg-1 propranolol,and equivalent distilled water respectively,one time a day,eight days in a row.After 30 min of last administration,the rats were anesthetized with ip intraperitoneal injection of chloral hydrate 350 mg· kg-1.The normal ECG was recorded after 20 min stable operation,and multi-point sc subcutaneous injection with 1 mg·kg-1 Iso to the rats of each group.Meanwhile,the rats of control group were injected with equal volume of normal saline,and recorded the change of T wave and J point of ECG in 20 min after the injection.At the end of test,blood sample were collected from femoral artery,and the contents of GSH-PX,SOD,and MDA in serum were measured.RESULTS The T-wave of ECG increased obviously after sc subcutaneous injection with Iso 5 min and 20 min.Compared with the control group,statistical difference existed,while the CITA dose groups reduced the T wave height.But the effect was weaker than the model group,and there was no significant difference.The J-point height of EGC decreased obviously,compared with the control group.The statistical difference existed,while the significant effect of 100 mg CITA against Iso,compared with the model group,the statistical difference existed.The content of serum MDA and GSH-Px increased significantly,compared with the control group,statistical significant existed.However,the activity of SOD in serum changed little,no statistical difference existed compared with the control groups.The 100 mg·kg-1 CITA and 5 mg· kg-1 CITA could obviously reduce the levels of isoprenaline rats serum MDA and GSH-Px,and statistical difference existed among groups compared with the model groups.CONCLUSION The CITA has protective effect on the injure caused by acute myocardial ischemia,and the effect may be related to the decreased myocardial contractility,expansion of coronary blood vessels,free radical generation reduced,and blocked lipid peroxidation reaction.
