乳腺癌组织中基质金属蛋白酶-9的表达及其意义

目的 :探讨基质金属蛋白酶 9(MMP 9)在乳腺癌患者病程进展中的临床意义。方法 :对 4 8例原发性乳腺癌患者癌组织石蜡标本 ,采用标准链霉菌抗生物素蛋白 过氧化物酶亲合免疫组化法检测肿瘤组织中基质金属蛋白酶 9的表达 ,分析其与临床病理特征间的关系。结果 :4 8例乳腺癌中MMP 9阳性表达率为 4 2 % ;本组患者中无腋淋巴结转移组、高分化组、无远处转移组及 5年生存组MMP 9表达水平低于有腋淋巴结转移组、低分化组、远处转移组及死亡组 ,差异有显著性意义 (P <0 .0 5 )。结论 :MMP 9和乳腺癌患者的转移和预后相关 ,此结果为临床建立新的乳腺癌预后预测因子提供了有价值的信息 ,并可用以指导临床治疗。

Expression of MMP-9 in Human Breast Cancer and its Implication

Objective:To investigate the clinical significance of matrix metalloproteiase(MMP) 9 in human breast cancer. Methods:The expression of MMP 9 in parafin embedded breast carcinoma specimens from 48 patients with primary invasive breast cancer who had undergone radical mastectomy or modified radical mastectomy from January of 1990 to Dcecember of 1996 were detected by the standard streptavidin biotin peroxides method,and its correlations to clinicopathologic features were analyzed. Results:Among 48cases,the positive expression rate of MMP 9 was 42%. MMP 9 expression was higher in patients with axillary lymph node metastasis than in those without metastasis ( P <0.05),and it was higher in patients with higher histological grades than in those with lower histological grades ( P <0.05).And MMP 9 expression was higher in patients with distant metastasis than in those without distant metastasis( P <0.05).Patients with higher expression of MMP 9 had a worse prognosis than those with lower expression( P <0.05). Conclusion:MMP 9 is correlated with metastasis and prognosis in breast cancer.These results provide a valuable clue for developing some new practical prognostic factors in breast cancer patients,which may be useful to direct clinic therapy.
