曲尼斯特在兔眼小梁切除术后抗纤维化的实验研究

摘    要:目的探讨曲尼斯特在实验性兔眼青光眼滤过术后抑制纤维瘢痕形成的价值。方法27只兔子(54眼)分为对照组、丝裂霉素C(MMC)组、曲尼斯特组,双眼均行标准小梁切除术。MMC组术中巩膜瓣下应用0.4mg/mlMMC,曲尼斯特组术后应用0.5%曲尼斯特滴眼液滴眼。分别于术后7天、14天、28天每组各处死兔子3只,摘除眼球、组织固定,用H—E、Masson三色染色后进行病理组织学检查。结果术后7天、14天、21天MMC组及曲尼斯特组眼压低于对照组(P〈0.05);而曲尼斯特组和MMC组眼压差异无统计学意义(P〉0.05)。3组功能性滤过泡生存率差异有统计学意义(P〈0.01);而MMC和曲尼斯特组差异无统计学意义(P〉0.05)。HE染色对照组术后7天成纤维组织在滤过道大量增生。Masson三色法光镜观察,对照组的新生胶原纤维排列有序且致密;而曲尼斯特组和MMC组的新生胶原纤维排列较紊乱.致密度低。结论曲尼斯特能抑制青光眼滤过术后滤过道纤维组织增殖,有效降低眼内压.维持滤过泡的形态和功能。

Abstract:Objective To evaluate anti-fibrosis effects of postoperative application of tranilast after experimental filtering surgery in rabbits. Method:Trabeculectomy were performed on twenty-seven rabbits (54 eyes). In treated group, 0.4 mg/ml MMC were used subscleral for 3 minutes; or 0.5% tranilast eyedrops were used after operation.
