电针对骶髓损伤后神经源性膀胱大鼠凋亡相关因子Bcl-2、Bax 的影响

摘    要:目的 观察电针次髎、中极、三阴交穴对骶髓损伤后神经源性膀胱(逼尿肌无反射型)大鼠膀胱组织中B 淋巴细胞蛋白-2(bcl-2)及Bcl 相关X 蛋白(bax)表达的影响,以探讨电针治疗该病的可能机制.方法 将SD 雌性大鼠40只,随机抽取10 只为空白组,其余造模成功后随机分为模型组、电针穴位组和电针对照点组,每组10 只.术后第15 天起电针穴位组、电针对照点组分别施以每日1 次治疗,治疗7 次后取膀胱组织行Elisa 法检测bcl-2 及bax 在膀胱组织中的表达.结果 电针治疗后各组膀胱逼尿肌组织中,模型组、电针穴位组和电针对照点组bcl-2 及bax 表达均较空白组高(P〈0.01),电针穴位组和电针对照点组bcl-2 表达较模型组升高(P〈0.01),bax 表达较模型组降低(P〈0.01),且电针穴位组疗效明显优于对照点组(P〈0.01).结论 电针次髎、中极、三阴交穴可提高骶髓损伤后NB 大鼠膀胱组织中bcl-2的表达、抑制bax 的表达.电针对骶髓损伤后神经源性膀胱的治疗作用可能是通过促进bcl-2、抑制bax 的表达以部分抑制膀胱平滑肌细胞凋亡,从而保护受损膀胱逼尿肌组织.

Abstract:Objective To explore the mechanisms of electro-acupuncture therapy by observing the effects of electro-acupuncture at Ciliao, Zhongji, Sanyinjiao acupoint on the expression of bcl-2 and bax in bladder tissues of rats with neurogenic bladder (detrusor areflexia type) after sacral spinal cord injury. Methods 10 SD rats were randomly selected as blank group from 40 SD female rats. The others were randomly divided into model group, electro-acupuncture group and control point group after modeled, with 10 rats in each group. The rats in electro-acupuncture group and control point group were treated from the 15th day after operation and lasted for 7 days. The expression levels of bcl-2 and bax in the bladder tissues were detected by ELISA. Results The ex- pression levels of bcl-2 and bax in the bladder detrusor tissues were increased in model group, electro-acupuncture group and control point group than those of blank group (P〈0.01). The expres- sion levels of bcl-2 were increased and bax were decreased in the bladder detrusor tissues in elec- tro-acupuncture group and control point group than those of model group (P〈0.01). The therapeutic effects of the electro-acupuncture group was superior to the control point group(P〈O.O1). Conclu- sion Therapeutic effects of electro-acupuncture of Ciliao, Zhongji, Sanyinjiao acupoint on the treatment of neurogenic bladder after sacral spinal cord injury may be played by improving bcl-2 expression, reducing bax expression and partially inhibiting the apoptosis of bladder smooth muscle cells, then contributing to the impaired bladder detrusor tissues.
