黄连温胆汤加减对代谢综合征大鼠胰岛素抵抗及脂肪细胞因子的影响

摘    要:目的:观察黄连温胆汤加减对膳食诱导代谢综合征大鼠胰岛素抵抗及脂肪细胞因子、瘦素、FFA、TNF-α的影响,探讨相关作用机制。方法:采用高脂高糖高盐饲料喂养建立MS大鼠模型,选择符合标准的MS大鼠随机分为模型组、非诺贝特组、黄连温胆汤高、低剂量组,相应药物灌胃4周,检测体重、血压、血糖、血清胰岛素、血脂、瘦素、FFA、TNF-α,计算lee’s指数和胰岛素敏感指数。结果:黄连温胆汤高剂量组和非诺贝特组体重、血糖、血清胰岛素、胰岛素敏感性指数、Lee’s肥胖指数、内脏脂肪体重比、TG、TC和LDL-C较模型组均下降(P<0.05);非诺贝特组HDL-C明显增高(P<0.05);仅黄连温胆汤高剂量组血压下降(P<0.05);3组大鼠瘦素、FFA、TNF-α均显著降低(P<0.05)。结论:黄连温胆汤加减对MS大鼠各代谢组分均有治疗作用,其机制可能与改善胰岛素抵抗和调节瘦素、FFA、TNF-α等脂肪细胞因子表达有关。

Abstract:Objective:To observe the effect of Huanglianwendan decoction on insulin resistance and adipocytokines, leptin, FFA, TNF - α in diet - induced metabolic syndrome rats, and explore the relevant mechanism.Methods: establish MS model rats by feeding high -fat -high -sugar- high -salt diet ,the rats which has satisfied the standard of MS model were randomly divided into four groups, which contained model group, fenofibrate group, Huanglianwendan dacoction high and low - dose group. Each group garage the corresponding drag 4 weeks, test weight, blood pressure, blood glucose, serum insulin, lipids, leptin, FFA,TNF -α, and calculate lee's index and insulin sensitivity index. Results : The body weight, blood glucose, serum insulin, insulin sensitivity index, Lee's body mass index,visceral fat weight ratio,TG,TC and LDL - C in Huanglianwendan dacoction high - dose group and fenofibrate group rats decreased obviously compared with model group( P 〈 0. 05 ) ;HDL - C in fenofibrate group was significantly higher( P 〈 0. 05) ;blood pressure in Huanglianwendan decoction high-dose group decreased only (P 〈 0. 05 ) ;the level of leptin, FFA and TNF - α in those three groups rats were significantly lower ( P 〈0. 05 ). Concluaion : Huanglianwendan decoction has curative effect on every metabolic components of MS rats, and the mechanism may be related to improving insulin resistance and regulate the expression of leptin, FFA,TNF- α and other adipocytokine.
