缺氧诱导体外培养的心肌细胞凋亡及检测方法研究

为探讨缺氧诱导体外培养的心肌细胞凋亡情况，并用不同方法检测，将体外培养的不同方法检测，将体外培养的新生大鼠心肌细胞置于95%氮气及5%二氧化碳孵箱中一定时间造成缺氧损伤的心肌细胞模型，分别用HE染色，电镜，TUNEL法，流式细胞仪技术检测凋亡发生的情况，结果：均观察到缺氧诱导的心肌细胞凋亡，认为缺氧一定时间可以诱导体外培养的心肌细胞凋亡；检测心肌细胞凋亡的方法，需结合形态学及定量方法全面分析。

A Study on Hypoxia Induced in Vitro Cultured Cardiac Muscle Cell Apoptosis and Its Detection Method

To probe into the hypoxia induced in vitro cultured cardiac muscle cells apoptosis, hypoxia damaged cell model was built via in vitro culturing of cardiac muscle cells of neonatal rat under 95% N 2 and 5% CO 2 of incubation box and kept it for a period of time. HE stain, electron microscope, TUNEL and flow cytometer technique were used respectively to detect the apoptosis. Results showed that cardiac muscle cell apoptosis induced by hypoxia had observed in above mentioned 4 methods. Suggested that hypoxia for a certain period of time may induce in vitro cultured cardiac muscle cells to develop apoptosis. To detect the apoptosis, it should combine with both morphological changes and quantitative method.
