摘    要:目的 研究抑癌基因p15 mRNA在肺癌组织中的表达,揭过p15基因的失活与肺癌的发生和其组织类型的相关性。方法 对病理确诊的93(非小细胞肺癌65,小细胞肺癌28)例肺癌病理组织切片,采用p15 cDNA探针以原位杂交组织化学方法检测p15 mRNA在肺癌组织中的表达水平。结果 p15 mRNA阳性表达于肺癌组织细胞质或细胞核中,在细胞质中可到较多的棕黄色颗粒,而细胞核内棕黄色颗粒相对少见。p15 mRNA的阳性表达率:非小细胞肺癌为46%(30/65),其中鳞癌46%(23/50),腺癌47%(7/15)。小细胞肺癌20%(5/25)。非小细胞肺癌p15 mRNA阳性表达率高于小细胞肺癌;高分化的鳞、腺癌阳性表达率高于同组中的低分化的鳞、腺癌(P<0.05)。结论 p15基因的失活与肺癌的发生密切相关,而且其失活程度与肺癌组织类型的恶性程度呈负相关。

Abstract:AIM To show the association of inactivation of p15 gene in lung cancers with pathogenesis and variation of lung cancer tissues by studying the expression of p15 mRNA in lung cancers. METHODS There were 93 cases, of which 65 were NSCLC and 28 were SCLC, confirmed by pathologicaly. The expression of p15 mRNA in pathological sections of lung cancer was examined using p15 cDNA and in situ hyhyidizatino histochemistry method. RESULTS In 93 lungcancers, positive expression of p15 mRNA was found located in the cytoplasm or nuclei, with quite a lot of brown yellow granules seen in cytoplasm, but only a few in nuclei. In NSCLC cases the positive rate of p15 mRNA expression was 46% (30/65), of which, lung squamous carcinoma and lung adenocarcinoma were 46% (23/50) and 47% (7/15) respectively, and the positive rate in SCLC cases was 20% (5/25). The positive rate of p15 mRNA expression was higher in NSCLC than in SCLC ( P <0.05). The positive rate in well differentiated lung squamous carainomas and lung adenocarainomas was higher than that in poorly differentiated ones ( P <0.05). CONCLUSION The results showed that p15 inactivation might be strongly related to pathogenesis of lung cancers and the degree of p15 inactivation might be negatively associated with the malignant degree of the lung cancer tissues.
