摘    要:目的动态观察脑缺血再灌注时鼠脑纹状体区细胞外液半胱氨酸(Cys)含量变化及丹参的影响,探讨Cys对缺血性神经元的损害作用。方法应用脑内微透析技术,采用高灵敏度的高压液相色谱-电化学检测手段,动态观察沙土鼠前脑缺血30分钟再灌注120分钟时,纹状体区细胞外液Cys的变化。结果前脑缺血30分钟时,细胞外液Cys浓度迅速升高,达缺血前的18倍。再灌注30、60分钟时,分别为缺血前的5倍和2倍,90分钟时基本恢复到缺血前的水平。丹参治疗组脑缺血时细胞外液Cys的含量较对照组低。结论Cys参与了脑缺血再灌注引起的损伤,而丹参可降低Cys水平,从而对缺血脑组织起到保护作用。

Abstract:Objective To investigate the association of extracellular fluid cysteine (ECF Cys) levels of striatum with cerebral ischemia and reperfusion as well as the effect of radix salviae miltiorrhizae (RSM) on the ECF Cys levels in the striatum with varying durations (10, 20, 30 min) of transient forebrain ischemia in gerbils. Method Microdialysis was used to sample the extracellular fluid in the right striatum during the 30 min forebrain ischemia followed by 120 min reperfusion. The microdialysate was measured by high performance liquid chromatography with electrochemical detection (HPLC ED). Results The ECF Cys levels were rapidly increased (18 folds) during the first 30 min after forebrain ischemia. After reperfusion, the ECF Cys levels increased 5 and 2 folds for the two reperfusion periods (0 30 min, 31 60 min), respectively. After the 90 min reperfusion, the ECF Cys levels returned to the levels before the cerebral ischemia. Although the ECF Cys levels in the RSM treated gerbils were also increased, they were significantly lower than those of the control ones. Conclusion The results suggested that Cys was involved in the pathophysiological processes of cerebral ischemia and reperfusion injury. RSM could have a neuroprotective role by the decreasing ECF Cys levels during cerebral ischemia and reperfusion.
