| 摘 要： | 目的体外合成小鼠CD40序列特异性shRNA，并构建连接于质粒表达载体，并进行序列分析，为开辟诱导免疫耐受的新途径提供实验依据。方法设计有小发夹结构的两条DNA序列。经退火成互补双链，再连接至pGenesil-1质粒载体，转化DH5a菌株，提取质粒行酶切鉴定后，进行测序分析。结果成功构建了CD40 shRNA质粒表达载体。结论CD40 shRNA质粒表达载体的成功构建为研究RNA干扰CD40基因表达，诱导免疫耐受奠定了基础。

| Abstract: | Objective To construct the plasmid vetor carrying CD40 shRNA and analyze the nucleic acid sequence for further searching new gene therapy method of inducing immunological tolerance . Methods Two DNA sequences containing small hairpin structure were designed and synthesized. The complement form was obtained by annealing and inserted into vector Pgenesile - 1 plasmid vector, and the recombinant plasmid was transformed into DH5a strain. Finally tbe plasmid identified by restriction enzyme was used for sequence analysis. Results The Pgenesile - 1 plasmid vector carrying CD40 shRNA had been constructed and the objective sequence had been obtained. Conclusions The construction of the plasmid vector carrying CD40 shRNA making a basis for the study of its inhibitive effect on inducing immunological tolerance. |
