“胆病从肝论治”在防治胆结石实验研究中的应用

目的:以"胆病从肝论治"为原则研究大黄灵仙胶囊防治胆结石的作用机制。方法:使用致石饲料诱发方法建立豚鼠胆囊结石模型,治疗组造模期间每日予大黄灵仙胶囊(制作成悬液)灌胃治疗,连续8周。观察比较治疗组和模型组豚鼠胆囊结石形成率,检测各组豚鼠肝脏病理和超微结构变化情况以及荧光定量PCR检测肝组织胆固醇、胆汁酸代谢相关酶的表达量。结果:模型组和治疗组豚鼠胆囊结石形成率分别是100%和50%,差异有统计学意义(P0.01)。治疗组豚鼠肝组织病变较模型组明显减轻,胆固醇7a-羟化酶、胆汁酸盐输出泵基因表达分别是模型组的2.08倍和2.57倍(P0.01)。结论:大黄灵仙胶囊具有较好的防治结石功效,作用机制与其保护肝组织以及在转录水平上调胆固醇、胆汁酸代谢相关酶的表达有关。"胆病从肝论治"在胆结石防治实验研究中具有重要的指导价值。
