微波对凡士林油纱条间歇灭菌的试验观察

目的:观察微波间歇灭菌对凡士林油纱条的灭菌效果。方法:将枯草杆菌黑色变种芽胞菌片放入10、20、30层凡士林油纱条的上、中、下层,微波(频率2 450MHz,功率650W),照射30、40、45、50min,于24~48h后再照射相同时间,每次照射后,取出菌片接种于无菌肉汤管中,37℃培养72h,观察有无菌膜生长,并测试温度和记录耗水量。结果:微波照射50min,经3次灭菌,可将各层凡士林油纱条的枯草杆菌黑色变种芽胞全部杀灭;30层油纱条照射45min,第2次灭菌,即可将枯草杆菌黑色变种芽胞全部杀灭;温度在75℃~85℃之间,耗水量为205~285ml。结论:30层内的凡士林油纱条经微波间歇灭菌3次,每次50min,可全部达到灭菌。

Observation of intermittent sterilization on vaseline gauze by microwave

Objective To observe the effect of intermittent sterilization on vaseline gauze by microwave.Method Three groups separately with ten,twenty and thirty layers of vaseline gauze were tested.Spore strips of Bacillus subtilis Var.niger were put into various layers in 3 groups,then irradiated by microwave(frequency:2 450MHz,power:650W) for 30min,40min, 45min and 50min respectively.They were irradiated again for the same period of time after 24～48h.After each irradiation,the strips were taken out and inoculated into the sterile broth,in order to observe whether the bacterial film was formed after the sterile broth had been incubated at 37℃ for 72h.Temperatures were tested and quantity of consumed water was recorded after each test.Results Spores of Bacillus subtilis Var.niger in the vaseline qauze of three tested groups were totally killed after the strips were irradiated for 50min by microwave and sterilized for three times.Spores of Bacillus subtilis Var.niger in the thirty layers of vaseline qauze were totally killed after irradiated for 45min with two times of sterilization.The tested temperature was between 75～85℃ and the quantity of consumed water was 205～285ml.Conclusion The vaseline gauze within 30 layers are totally sterilized after irradiated for three times(50min each time) of intermittent irradiation by microwave(frequency:2 450MHz,power:650W).
